Objective Preparing oncolytic adenovirus H101as a vector plasnid and inseringCD147shRNA to construct H101-shRNA, then assay the ability of tumor cells’proliferation and proteins(MCT, MMP) activity associated with invision andmigration, and determine the CD147therapeutic effect on colorectal carcinoma.Methods CD147shRNAs are cloned into shuttle plasmids pDC316-EGFP-U6which are then cotransfected with an Ad genomic plasmid (H101) into293cells, bothtwo were catalyzed by Cre or FLP to construct H101-shRNAs. Then, we use therecombind adenovirus plasmid to infect cell line HCT8with nomal P53and HCT15with inactive P53, mesure the cell proliferation and assay proteins activity linked withinvasion and migration.Results The recombind recombind adenovirus can proliferate in p53mutationtumor cell and affect the invision and migration of the tumor cell by inhibitingCD147expression. However, it has no effiction on nomal P53tumor cells.Conclusions The recombind oncolytic adenovirus H101-CD147shRNA can speciallykill the tumor cells by its toxiferous proliferation and inhibit proteins activityassociated with invision and migration.
