Objective:Form the mechanics perspective of cell biology, in vitro simulation of dynamic mechanical to stimulate to explore mature adipocity apoptosis,adiponectin and the coupled receptor of adiponectin (AdipoRl、AdipoR2),visfatin mRNA are released on the impact of the expression for control simple obesity bases on theory and experiment.Methods:1. We used the28-day-old male Sprague Dawley Rat as experimental material,sterile acquisition the around of epididymis’adipose tissue,isolation and culture mature adipocity by using of enzyme digestion. Through morphology observe,inspection mature adipocity indicator marker gene adiponectin by Oil Red O staining method and RT-PCR method to explore primary cell authentication, and with constant temperature culture device to load dynamic mechanical for cell.2. Digestion separation the adipose tissue of30male SD rats epididymis around to get mature adipocity,then random divide all cell into five groups—0R、60R、90R、120R、150R. After24hours synchronization,according to homologous group to load different stimulate,times stimulate every day,stimulate30min every time,the spacing interval is12hours,continue doing3days,6times in totally. After that,collecting cell to abstract DNA,total RNA, separately using DNA gel Electrophoresis to authenticate DNA Ladder,observe the phenomenon of apoptosis.3. Successfully detached the destined elements and cultured the well-ripe fat cell in vitro. Also the oil red O chromosome was testified positive.Through RT-PCR method the result of fat cell trait also suggested positive. Besides,this result is accurate and the data strengthens the confidence that this practice can be trusted.2. Through the DNA gel electrophoresis,become conscious of the phenomenon of DNA Ladder, and it have a obvious phenomenon which is strength intensity dependence, the stimulation strength become stronger,at the same time the phenomenon of apoptosis become more obvious.RT-PCT semi-quantitative analyze Bax、Bcl-2genes mRNA expression change in every rotate speed group,Bax genes mRNA expression change expression level ascend along with the stimulation strength become stronger, and every group have obvious difference (P<0.01),Bax genes mRNA expression change expression level have a slight lessen phenomenon, but it didn’t have obvious difference(P>0.05) 3. RT-PCT semi-quantitative analyze APN、AdipoRl、 AdipoR2、Visfatin genes mRNA expression change in every rotate speed group,①APN genes mRNA expression:Compare with0Rgroup,60R group and90R group APN expression level is higher than OR group,there is the phenomenon and it is obvious difference (P<0.01) had up-regulated trend;compare with90group,120R group and150group APN expression level is lower than90group (P<0.01),there is the phenomenon and it is obvious difference,had down-regulated trend;②AdipoRl genes mRNA expression:compare with OR group,60R group and90R group APN expression level had up-regulated trend,60R group is higher than OR group,but it didn’t have obvious difference.(P>0.05),90R group is higher than OR group, it had obvious difference (P<0.05). Compare with90R group120R group and150group AdipoRl expression level is lower than90group (P<0.01),it had obvious difference, had down-regulated trend;③AdipoR2genes mRNA expression: compare with OR group,60Rgroup and90R group AdipoR2expression level had up-regulated trend,60R group is higher than OR group, but it didn’t have obvious difference.(P>0.05),90R group is higher than OR group, it had obvious difference(P<0.01). Compare with90R group120R group and150group AdipoR2expression level is lower than90group,it had obvious difference (P<0.01), had down-regulated trend;④Visfatin2genes mRNA expression:Compare with ORgroup,60R group and90R group Visfatin expression level is higher than OR group, there was the phenomenon and it was obvious difference (P<0.01) had up-regulated trend;Compare with90group,120R group and150group Visfatin expression level is lower than90group, there is the phenomenon and it is obvious difference (P<0.01), had down-regulated trend;Conclusion1.Under the available lab condition, and through the toughest struggle to construct and imitate the situation when ripe fat cells are pressed within body,finally we have built a model which would show how massage would work the magic, and thereby lay a profound foundation for treating pure obesity by dealing with fat cells.2. External simulation of massage would stimulate the dying processes of ripe fat cell. By changing the pair of Bax and Bcl-2with Dynamic mechanics is one of the efficient ways performed by fat-lose-oriented massage.3. People who suffer obesity also bear a danger of serious IR,thus by applying certain and proper massage would escalate the amount of one’s adiponectin which would minimize IR so that one can lose weight.4. Overpoweringly applying massage to one would reduce the rate of adiponectin,which suggests it is possible to start some negative feedback and restrain the expression level of adiponectin as well as its receptor and Visfatin. However specific mechanisms are still remaining to be testified.
