Res To Add Bfgf Cell Proliferation Of Mice B16 Melanoma Cells And Melanin Formation

Objective:Some methods of culture mouse melanoma cell B16were used in established cell model.The methods were added different concentration of bFGF in different time.Then observe the RES on melanoma cells’ proliferation and melanoma cells’ synthesis of melanin in the cell model.Methods:1.The establishment of a cell model:to cultivate mouse melanoma cell B16in laboratory and intervene the cells in pharmaceutical culture-medium with five levels of concentration prepared with the bFGF,contrasting with the control group in blank culture-medium. After24hours,48hours,72hours, to detect the conditions of melanoma cells’ proliferation, mouse melanoma cells’ synthesis of melanin with MTT assay, NaOH fragmental assay.Then, Choose the drug concentration and time about make the most cells’ proliferation and the largest amount of melanin synthesis to establish a cell model.2.The RES influence on the cell model:Three levels (25mu g/ml,50mu g/ml,100mu g/ml) of concentration prepared with the RES and hydroquinone were be used to to intervene on the cell model.Then to detect the conditions of melanoma cells’ proliferation, mouse melanoma cells’ synthesis of melanin with MTT assay, NaOH fragmental assay respectively.Results:1.The establishment of a cell model:Among five levels of concentration prepared with the bFGF, it is the level of10ng/ml and the of48hours which has fine boost effect on cell proliferation and melanin synthesis. 2.The RES influence on the cell model:The best level of concentration prepared with the RES is50μg/ml to effects after the index.Conclusion:The RES, which is safe and non-toxic crystal, has an excellent inhibiting effect on cell proliferation and melanin synthesis. Therefore, it is good choice for removing freckles and face-whitening, and can be applied in the domains of chromatodermatosis’treatment and face-whitening.
