Based on the acupuncture formula for inducing labor optimized by our research group (EA at bilateral LI420min, plus EA at bilateral SP65min, totally retaining25min, sparse-dense wave,2/50Hz), the purpose of this research is to study its effect on cervical ripening in late-stage pregnant rats, so as to provide certain experimental evidence for revealing its scientific connotation.ABCTRACT Objective:To study the effect of electroacupuncture (EA) at points of an optimized traditional acupuncture formula that induces labor(EA at bilateral LI420min, plus EA at bilateral SP65min, totally retaining25min, sparse-dense wave,2/50Hz) on cervical ripening and its mechanism from the perspective of molecular biology, by observing cervical ripening related indexes in late-stage pregnant rats.Methods:Twenty pregnant Wistar rats were randomly divided into EA group (n=10) and model group (n=10), A normal control group included10non-pregnancy female rats. Rats in EA group received EA at points of an optimized traditional acupuncture formula that induces labor. EA was given on bilateral Hegu (LI4) for20min and then on bilateral Sanyinjiao (SP6) for5min with2/50Hz sparse-dense waves. No EA on model group and normal control group. ELISA method was used to detect the contents of prostaglandin E2(PGE2),interleukin8(IL-8) and matrix metallopeptidase9(MMP-9)in cervical tissue.Results:1.Comparison of PGE2contents in rat’s uterine among three groupsCompared with normal control group, PGE2contents in model group were increased significantly (79.146±6.3529pg/ml versus69.219±7.7706pg/ml, P<0.01). Compared with model group, PGE2contents in EA group were increased significantly (88.353±6.0133pg/ml versus79.146±6.3529pg/ml, P<0.01).2.Comparison of IL-8contents in rat’s uterine cervical among three groupsCompared with normal control group, IL-8contents in model group were increased significantly (45.363±5.8375pg/ml versus27.949±7.2539pg/ml, P<0.01). Compared with model group, IL-8contents in EA group were increased significantly (52.928±9.4603pg/ml versus45.363±5.8375pg/ml, P<0.05).3.Comparison of MMP-9contents in rat’s uterine cervical among three groups Compared with normal control group, MMP-9contents in model group were increased significantly (4.44341±0.96516ng/ml versus3.18698±0.82122ng/ml, P<0.01). Compared with model group, MMP-9contents in EA group were increased (5.34747+0.91174ng/ml versus4.44341±0.96516ng/ml, P=0.05).Conclusion:Optimized traditional acupuncture formula for inducing labor can increase the contents of PGE2, IL-8, MMP-9in cervical tissue of late-stage pregnant rats, so as to promote cervical ripening. This study explained the mechanism of EA in promoting cervical ripening from the perspective of molecular biology.
