Objective:To investigate the effect of Huannao Yicong Formula on Learning and memory capacity, pathological form the hippocampus, Brain oxidative stress indicators superoxide dismutase (SOD), malondial dehyde (MDA), glutathione (GSH), glutathione peroxidase (GSH-PX) content, inflammatory factor, interleukin-1(IL-1) and interleukin-6(IL-6) levels and Hippocampal CAl area β-amyloid peptide and TTBKl positive expression of senescence accelerated mouse prone8(SAMP8).Methods:The rapid aging mice (SAMP8, male) for experimental animals, totally of50,Randomly divided into5groups:Model group, Positive drug group (donepezil hydrochloride,0.65mg/kg),large dose group of HNYCF treatment (21.84g crude drug per kilogram), middle dose group of HNYCF treatment (10.92g crude drug per kilogram), low dose group of HNYCF treatment (5.46g crude drug per kilogram), Each group of10only.Normal aging mouse(SAMRl) as normal group. Fill the stomach to medicine12weeks. Model group and normal group give equal distilled water. After the drug, the Morris water maze experiment of mice behavior changes, Anesthesia executed mice. Take the brain tissue, part of the tissue put in4%neutral formaldehyde fixed, Pathological conventional slice, HE dyed, Observe the hippocampus of mice pathological form. Part of the tissue cryopreserved, homogenate. Application colorimetric method to detect the activity and contents of SOD、MDA、GSH、 GSH-PX, use the radioimmunoassay to measure the contents of IL-6and IL-1in hippocampus and the cortex, Immunohistochemical method (color image processing system and half quantitative analysis) observation of hippocampus CAl area β-amyloid peptide and the expression of Tau protein kinase (TTBK1), Statistics of each group of positive expression of integral optical density (IOD) values.Results:1Water maze experiment results:Group compared with model, HNYCF can increase the number of SAMP8mice swimming through the platform, extend the mice in the fourth quadrants journey and time (P<0.05);2Mice hippocampus observed after HE staining:Model group mice hippocampus CA1zone cells is sparse, Arrange a disorder, Can see cell degeneration and necrosis and Hyperchromatic nuclei and pyknosis; Compared with model group, Dosage groups is more compact and neat arrangement of hippocampal CA1area cell, the nucleus and the cytoplasm is clear, less free denaturation and necrosis.3Mice brain oxidative stress indicators:Compared with normal group, Model group mice hippocampus and cortex in the brain tissue SOD and GSH-PX activity decreased, the GSH content decreased significantly Statistically significant difference (P<0.05, P<0.01), MDA levels increased significantly, Statistically significant difference (P<0.05, P<0.01); Compared with model group, drugs can increased the activities of SOD and GSH-PX in hippocampus and cortex, and the content of GSH,reduce the content of MDA, Statistically significant difference (P<0.05, P<0.01).4Mouse brain tissue inflammatory factor, interleukin-1(IL-1) and interleukin-6(IL-6) levels: Compared with normal group, Model group mice brain interleukin1(IL-1) and interleukin-6(IL-6) content increased, Statistically significant difference (P<0.05, P<0.01); Compared with model group, Each dosage group of IL-1and IL-6levels were lower, Statistically significant difference (P<0.05, P<0.01).5Immunohistochemical and IOD results:SAMP8hippocampal CA1area TTBK1positive expression of model group was obviously increased, β-amyloid deposition increase. Compared with model group. Each dose group TTBK1and β-amyloid peptide positive expression is significantly reduced. Application of computer image analysis system and half quantitative calculation IOD values significantly reduced, Compared with model group difference was statistically significant (P<0.05, P<0.01).Conclusion:HNYCF by inhibiting SAMP8brains oxidatie stress and in inflammatory reaction, Reduce brain tissue deposition of β-amyloid and Tau protein, improve the hippocampus and the pathological changes of the cortex. And then improve SAMP8mice space recognition and the ability of learning.
