Research Purposes: Stress stimulation is one of the most basic stimulationduirong bone remodeling process Appropriate stress stimulus can promote bone cellgrowth, differentiation and maintain function, but the excessive or duration longmechanical force to bone cell can lead damage, the mechanism is bone cell can responseto stress stimulus signal and convert to biological chemical signal, OPG, nuclear factorκ B receptor activation factor and its ligand (OPG/RANK/RANKL). Bone metabolismsignal path is one of stress sensitive pathways. OPG/RANK/RANKLis the key to keepdynamic balance of bone absorption and bone formation. In addition, the hormone canalso affect bone metabolism signal path, parathyroid hormone is the important hormoneto regulation bone metabolism signal pathway. Continuous movement stress canenhance hormone level inevitably, whether stress and hormone can form stimulation onbone. Therefore, this experiment to study the law of OPG、RANKL during jumpingexercise, and the change rule of parathyroid hormone on bone metabolism signal, fromgene level to study bone metabolism, provided the evidence for other studies.Research Methods: This experiment is that rats start six weeks of jumpingexercise in the cage with semi-automatic adjustable voltage, after exercise, killed ratsand taken the blood and the tibia. HE dyed slice to observe the inner change of bonetissue, ELISA method was used for determining the content of serum PTH, Real-timePCR was used for determining the level of OPG、RANKL mRNA of tibia tissue,Analysis the change regulation of OPG, RANKL.Research Results:1、HE dyeing section, fiber arrangement began to disorder, the number of bonelacunar increasing gradually, bone lacunar broadening gradually, bone trabecular loosing, the area of osteolysis increasing gradually with the load and time of jumpingexercise increasing.2、Control group two weeks, four weeks, six weeks rats, parathyroid hormone haschanged,has on statistical significance(P>0.05). Jump group two weeks and four weeksand six weeks,parathyroid hormone has changed, has difference and statisticalsignificance(P<0.05).Jump burden group two weeks, four weeks, six weeks, parathyroidhormone has a very significant change, has statistical significance(P<0.01).Jump burdengroup are higher than those of control group, jump group.3、Control group, jump group, jump burden group of two weeks rats OPGmRNAexpression gradually decreasing, with significantly difference, has statisticalsignificance(P <0.01); control group, jump group, jump burden group of four weeksrats OPGmRNA expression first increases and the reduce, with significantly difference,has statistical significance(P <0.01); control group, jump group, jump burden group ofsix weeks rats OPGmRNA expression first increased significantly after reduction,significantly difference, has statistical significance(P <0.01). Control group, jumpgroup, jump burden group of two weeks rats RANKLmRNA expression has slightincrease, with significantly difference, has statistical significance(P <0.01); controlgroup, jump group, jump burden group of four weeks rats RANKLmRNA expressionfirst increases and then reduce, with significantly difference, has statistical significance(P <0.01); control group, jump group, jump burden group of six weeks ratsRANKLmRNA expression increases gradually, which significantly difference, hasstatistical significance(P <0.01). control group, jump group, jump burden group of twoweeks rats OPG/RANKL ratio gradually decreasing, with significantly difference, hasstatistical significance(P <0.01); control group, jump group, jump burden group of fourweeks rats OPG/RANKL ratio increase gradually with significantly difference, hasstatistical significance;(P <0.01), control group, jump group, jump burden group of sixweeks rats OPG/RANKL ratio first increased significantly after reduction, withsignificantly difference,has statistical significance(P <0.01).4、 PTH and OPG, RANKLhas significant correlation, has statistical significance(P <0.01). Research Conclusion:1、The heavy load of jumping exercise of cannot achieve bone-invigorating, butaffect the normal bone metabolism, destruct the dynamic balance of bone resorption andbone formation, bone tissue was excessive absorption, cause bone micro damage andfracture.2、Stress stimulation caused hormone level changes, the parathyroid hormone ofdifferent groups of rats is different, parathyroid hormone increased with the load ofjumping exercise increasing, the big strength exercise may cause parathyroid hormoneincreaseing dramatically.3、The RANKL、 OPGmRNA expression and OPG/RANKLratio of rats ofdifferent load is different. OPGmRNA expression first increaseand then reduce,RANKlmRNA express has been a trend of increase, OPG/RANKLratio first increasesand sharply declining the load of jumping exercise increasing.4、Under parathyroid hormone effect, RANKLmRNA expression continuallyincreasing, on the contrary, OPGmRNA expression has the trend of decline with theincrease of PTH. The result shows that long time and continuously PTH maybe increasebone resorption.
