Research And Design Of Device For Temperature Calibration On Polymerase Chain Reaction Instruments

Nowadays, PCR instrument becomes important analytical equipment in the fieldof molecular biology with the development of life science and technology. PCRinstrument, also called the polymerase chain reaction instrument (Polymerase ChainReaction Instruments, hereinafter referred to as PCR system), is such an apparatusthat provide the specified temperature to guarantee DNA to copy itself. PCRinstrument realizes DNA degeneration and replication through thermal cyclingtemperature control. So our research of methods of testing is brand new and effective,meanwhile the device for PCR instrument for temperature calibration is designedreferred to the calibration specification. The thesis includes the following three parts:Firstly, it outlines the development of PCR instrument and relevant calibrationmethod at home and abroad, also the meits and demeits of PCR temperaturecalibration devices in being is summarized. We develop a wired and a wireless devicewhich can detect the temperature control of PCR instrument effectively and accurately,meanwhile we introduces its design and structure.Secondly, NTC sensor and its dimension are fixed by analyzing the temperatureenvironment of PCR in detail. USB communication is applied in the wired device,while the technology of Zigbee radio frequency is adopted in the wireless device. TheDesign of test system hardware circuit is completed. The design of computer softwareand system software, gives the system client software to send and receive data format,and details of the operating system software. The system’s hardware circuit isdeveloped; it provides electric schematic diagrams for every hardware module. Itdesigns the system’s software including the bottom software and the PC operatingsystem software, which is detailed.Thirdly, it sets up special temperature calibration device, which solves theproblem of system calibration, and validated the device performance. The metrologydevice achieves good performance including the system’s accuracy, repeatability,stability, uncertainty of the indication error etc. The experiment proves this system hasgood reliability, can be competent for temperature calibration of PCR instrument. Ourdevice made up the defects of existing device, and filled the blank in domestic on themarket.
