Oocyte and embryo could not benefit from the maternal antioxidant protection when divorced from the donor body, ROS could restrain oocyte maturation and emryo development. This is a restrict factor of in vitro culture. Epi-gallocatechin-3-gallate(EGCG) is a natural non-toxic antioxidant and an ideal added ingredients of in vitro culture system. In this study, rabbits were used as animal model, and different levels of EGCG were used to study its effects on IVM in order to improve the effects of in vitro culture and improve the in vitro culture system of rabbits.And then, the significance of BMP-15and FSHR genes expression in oocyte maturation were also investigated by the semi-quantitative RT-PCR.We researched the effects of BMP-15and FSHR genes on IVM in order to study its biological function on rabbits. The results were as follows.1. EGCG were added from0to25μM in TCM-199foundation culture systems. Discovered that the IVM rate improved along with the EGCG adding concentration increased. Adding15μM and20μM EGCG could improved the IVM rate significantly (P<0.05), but when EGCG added excessly,the IVM rate reduced obviously.There was no significant different effect between adding15μM and20μM for infancy rabbits(P>0.05); but the effect of adding20μM was better than adding15μM significantly for adult rabbits(P<0.05).2.In general, the IVM rate of adult rabbits was more than infancy rabbits(P>0.05); while,15μM or20μM EGCG were added in TCM-199foundation culture systems, the IVM rate of infancyt rabbits was more than adult rabbits(P>0.05).3.The BMP-15and FSHR genes were discovered that they had different expressions in the process of oocyte maturation by the semi-quantitative RT-PCR. BMP-15mRNA were expressed at low levels in immature oocytes, the expression increased gradually along with the time of maturation and peaked at12h of IVM.The expression had been maintain highly until24h of IVM, and since then,declined. These coincided with the time of cumulus cells expansion,so the expression pattern of BMP-15suggested that it might be important in cumulus cells expansion. While,FSHR mRNA were expressed at high levels in immature oocytes.There was a higher expression at0h and6h of IVM,and since then declined gradually.These may related with that oocytes needed more FSH at early maturation time,and it suggested that FSHR had important effects in oocytes maturation.4.The expressions of BMP-15and FSHR genes changed oppositely in the process of oocyte maturation,BMP-15gene controled negatively and restrained the expressions of FSHR gene.
