Actinomycete is a very important microbial resource of economic, medical and industrial application. However, with the increasing of the repetition rate of the known compounds isolated from the ordinary environmental actinomycetes, as well as the probability of discovered new active natural products are getting smaller and smaller. So, our research objectives aim to study microorganisms in extreme environments. Halophilic actinomycetes are a special group which living in the extreme high-salt environment, it is believed that a great metabolic diversity and biotechnological potential has been found in halophilic and halotolerant microorganisms.In this study, halophilic actinomycetes used in our research are isolated from salt lake in Xinjiang. To begin with, we selected30strains of halophilic actinomycetes of the anti-tumor activity and PKS-I gene on basis of previous work of our laboratory. Furthermore, we made use of little fermentation to incubate30strains of halophilic actinomycetes by means of three different broth mediums.The fermentation broth was extracted and concentrated to obtain the crude extract, in combination with TLC chemical diversity, we selected from the10strains of halophilic actinomycetes and optimal medium for fermentation. Finally, the10strains of halophilic actinomycetes amplified fermentation, the crude extract was isolated and purified of preliminary planning, we chose to YIM90615for the target by means of HPLC and TLC multi-level chemical screening, combined with comprehensive comparative analysis of the1D-NMR data. Strain of YIM90615is provided with high activity and stable metabolite.Following,150L broth cultures of YIM90615were harvested by extracting with an equal volume of EtOAc. We got25compounds by making use of a variety of isolation and purification methods to isolate and purify the crude extract. Based on the data of1H-NMR of25compounds, as well as the13C-NMR, MS, DEPT, HSQC,1H-1H COSY and HMBC data analysis of the parts of compounds, we identified the structure of19compounds, including the following types of compounds:5anthraquinones,3 isobenzofurans,2pyrimidines,2adenosines,1cyclo-(Tyr-Leu),1p-hydroxybenzaldehyde,13-indolecarboxylic acid,1p-hydroxyphenylacetamide,1N-(2-hydroxypropanoyl)-2-aminobenzoicacid amide,12’-deoxyadenosine,1tryptamine. There are5new compounds, including2anthraquinones and3isobenzofurans. The anthraquinones and isobenzofurans are isolated from halophilic actinomycetes at first time, as well as the isobenzofurans at first is isolated from actinomycetes.Recent studies show that halophilic actinomycetes have some special metabolites, which could produce a variety of different types of new compounds. Halophilic actinomycetes can be used as an important source of new natural products, and provide a new direction for the research and development of new drugs in the future.
