Leaves of mature plant of resurrection plant Boea hygrometrica (Bunge) R.Br. weredivided into fresh group, slow dehydration group and acclimated group to investigate thedesiccation-tolerance in the metabolimics levels represented by the changes of metabolismduring desiccation and subsequent rehydration. Meanwhile IAA, SA and JA three endogenoushormones and Vc, Ve two kinds of antioxidants were measured, and made a DAB and NBTstaining test to verify the tissue localization of over-hydrogen peroxide and superoxide anion,further research dehydration process of recovery in specific metabolite changes.First, The extracts of Boea hygrometrica using methanol solution (method A) andmethanol-chloroform-water solution (method B) was analyzed by gas chromatography-massspectrometry (GC-MS) in terms of the total numbers of chromatographic peaks, extractionefficiency, retention time and the peak stability. Determine to use the method methanol-chloroform-water law as an extraction method.Using methanol-chloroform-water extraction method as the extraction method for thedetection, to analysis the metabolites differences among different treatments. Metabolitesdifference mainly in carbohydrates, sucrose, Mannose, amino acids, includes tryptophan,Aspartic acid, organic acids, includes malic acid, butenedioic acid, acetic acid, ethanedioic acid,propanoic acid, pentanedioic acid, trihydroxybutyric acid and benzenedicarboxylic acid. Among them,pentanedioic acid, trihydroxybutyric acid and benzenedicarboxylic acid have not been reported. Whileseveral kinds of hydrocarbons, such as silanol, butane and octadecyne, have not been reported.With the dehydration, only the acclimated group shows that IAA, SA and JA in thethree groups show a downward trend, the real reason need further research.Hormone measurement results show that with dehydration progresses, IAA shows adownward trend only in the acclimated group. In fresh group and slow dehydration groupshouws a stable level. SA and JA in the three groups showed a downward trends.The result of Vc and Ve shows that the acclimated group’s Vc and Ve concentrationlevels are higher than the fresh group and slow dehydration group, indicating that after slowdehydration the plant has become the leader of the grass plants more resistant oxidizing materials, have a stronger antioxidant capacity, you can protect your organization reducestress damage.DAB and NBT staining test result shows that the slow dehydration damage lighter thanrapid dehydration. The hydrogen peroxide and superoxide anion content of acclimated groupis lesser than others. The acclimated group has stronger ability to resistant to oxidative stress.
