Phase contrast microscope is an effective method for observation of phase objects, it can image clearly for the biological tissue slice which is thin and not dyeing. And significantly, it has the characteristics of high resolution, high contrast.According to the characteristics of the biological tissue samples studied in this paper, that is a highly transparent phase, the phase contrast microscope imaging principle is analyzed detailly, so we choose the right phase contrast microscope as a research tool. Further discussed the Mie theory, mathematical morphology combined, we extract tissue granularity distribution through the disk structure element of mathematical morphology, and combine the granularity distribution with Mie theory. Finally we established a new model of the tissue optical properties, that can calculate and measure the optical properties of tissue, including the absorption coefficient, scattering coefficient and reduced scattering coefficient, mean cosine of scattering angle, and scattering phase function. In this paper, we first studied optical properties of rat tissue, and many aspects compared, the same tissue, between different tissues, between different magnification, were compared. This paper aims to find the relationship between the tissue optical properties. According to establishing model, the optical properties of human skin were described, and the optical properties along with the change of wavelength characteristics of the tissue can be know. By comparing the normal and cancerous tissues of human stomach, we found that the scattering characteristics of cancerous tissue is higher than normal tissue, this feature can be used to distinguish normal tissue and tumor tissue. The significance of this paper lies in using phase contrast microscope, collect and phase contrast images, model characterization of tissue optical properties parameters, these parameters have important significance.
