Objective:To investigate the effects of estrogen and estrogen receptor expression in the liver bypartially hepatectomy after total hepatic denervation, and to explore the mechanism of hepatic nerve inliver regeneration.Methods: Male SD rats (n=42) were stochastic divided into2groups: partially hepatectomy(PH), and partially hepatectomy after total hepatic denervation(PHT). Rats in each group were killed at12h,24h,36h,48h,72h,120hand168h after operation respectively, cut the rat liver organization.UseImmunohistochemical method to detect the expression of two estrogen receptors (ERα, ERβ) and theP450;use real-time quantitative (RT-PCR) method to detect the expression of ERαmRNA,ERβmRNA andcytochrome P450enzyme mRNA in rat regeneration liver cells.Results:(1)The experiment successfully build the rat2/3hepatectomy regenerating liver modeland complete denervation regenerating liver model.(2)Rat regenerating liver ERαexpression: Comparedwith PH group,the amount of ERαexpression in PHS group rats is increased at12h~168h afteroperation,the expression at36h~120h is increasing,samilary to the PH group rats and return to normallevel immediately,but significantly higher than PH group rats(P<0.05),the rat regenerating liver’s ERαexpression of PHT group reached peak at168h after operation;(3)Rat regenerating liver ERβexpression:Compared with PH group, the amount of ERβexpression in PHS group rats is increased at12h~36h afteroperation,the expression skills is weaken after operation,increased at168h after operation, and increasedsignificantly at24h,36h,120h,168h after operation(P<0.05)(.4)Rat regenerating liver P450expression:Compared with PH group, variation of the expression levels of the two groups of rats are moreconsistent,the expression levels of PHT groups is decreased at36h~120h, and have significant differenceat24h,48h,72h,120h after operation(P<0.05); after operation36h,168h expression is higher,at168hreach the peak.(5)Rat regenerating the liver cells ERαmRNA expression: Compared with PH group,thePHT groups’ ERαmRNA expression levels rised and reachd the peak, the expression was significanglydecreased at48h~120h after operation(P<0.05),the strongest expression ability is at24h and48h afteroperation..(6)Rat regenerating the liver cells ERβmRNA expression results: Compared with PH group,the PHT groups’ ERβmRNA expression level is significantly reduced at12h~72h after operation(P<0.05),the expression was significanglyincreased at120h after operation(P<0.05).(7)Rat regenerating the livercells P450enzyme mRNA expression results: Compared with PH group, the expression was significanglyincreased at12h~72h after operation(P<0.05),at120h was decreased,the expression at168h has thesignificant difference(P<0.05).Conclusion:(1)Total hepatic denervation can promote liver regeneration at48h-120h afteroperation.(2)The regulate mechanism of total hepatic denervation to rat remnant liver regeneration mayby promoting estrogen receptor α expression levels、controling estrogen receptor β expression levels andenchancing the body P450activity to regulate the body metabolism, promote the recovery of liverregeneration.
