With a variety of powerful active enzymes, Aspergillus niger is a filamentous fungus, isan internationally recognized security strains used in the food industry.In Aspergillus niger, YY-01as the original strain, the use of chemical reagents-ethylmethane sulfonate (EMS) as a mutagen treatment to obtain pectinase yield mutant strains.Transparent circle first screening selected transparent circle colony diameter ratio greatlyimproved mutants, and then re-screened by solid fermentation method, a yield pectinasemutant YY-21final screening. This strain after five consecutive generations slant culture,stable production pectinase genetic characteristics, its enzyme activity more than reach the130U/mL of3.6times higher than the original strain.Single factor and orthogonal experiment method of Aspergillus niger mutant strainYY-21solid-state fermentation conditions were optimized. Experimental results show that theAspergillus niger mutant strain YY-21, optimal culture conditions: medium inducer added inan amount of0.4g, and the initial pH of6, and the amount of inoculation of1×107(a/10gdry basis) extraction time was120minutes.Single factor and orthogonal experiments on juice clarification select the optimalconditions, the optimum conditions for633.32U/ml of active enzymes,90min,48°C, pHvalue of4.4, Clarity reached92.8%, after clarification of fruit juice containing Vc0.7mg/Lmagnesium ion content9mg/L, calcium content is15mg/L.
