14-3-3protein is a kind of rich content of highly conserved acidic protein, exists in eukaryotic cells. The14-3-3protein is found in the process of Moored isolated from bovine brain protein in succession, and later found in many eukaryotes.the14-3-3protein involved in Many of cells life activities, has important function in the cell,Mainly in the proteins consist in eukaryotic cells.14-3-3proteins first is Moored, etc the signal transduction, cell cycle, apoptosis, enzyme activity, etc have important function in the process of adjustment.Antheraea pernyi is a kind of is developing wild ilk-producting insect, which belongs to the Lepidoptera Saturniidae Antheraea. In addition to as raw materials for the textile industry, there are high medicinal and edible economical values.Using the method of molecular biology and bioinformatics study of tussah14-3-3protein, The major results are summarized as follows:1、The cDNA sequence of14-3-3protein,1220bp in lenth,an open reading frame of744bp, encoding247amino acids, the molecular weight size28.1KDa and isoelectric point of4.9.2、The ORF was amplified by RT-RCR, ligated to PET-28a(+) expression vector, and turned into escherichia coli BL21(DE3),inducted by IPTG under different concentrations,Through SDS-PAGE electrophoresis and Western blot analysis, i the inducing fusion proteins was successfully expressed. to increase the use of affinity column chromatography purification of the recombinant protein, this will provide the foundation for further study on eukaryotic expression and function of14-3-3proteins.
