In a pot experiment,the top fresh weight, accumulated amount of dry matter, the content of main composition of the stalk and the photosynthetic characteristics were ascertained, the effects of sucrose synthase, β-1,3-glucanase activity and IAA,ABA content on fiber development of the stalk in Autotetraploid and Diploid of Triarrhena lutarioriparia L.Liu was studied. Mean while, Cellulose synthase gene fragment was cloned, and then analyzed by bioinformatical methods.The main research results in this experiment are as follows:1Some physiological characteristics of Autotetraploid and Diploid of Triarrhena lutarioriparia L.Liu are ascertained. Net photosynthetic rate in Autotetraploid and Diploid of Triarrhena lutarioriparia L.Liu show the same changing trends, it reaches the maximum value at vegetative growth stage Ⅱ, respectively31.88μmol·m-2·s-1,27.07μmol·m-2·s-1. The top fresh weight of leaves and stalk rapidly increased during early growth stage. Compaerd with growth stage I, the height, diameter of the stalk and accumulated amount of dry matter in the stalk were initially increased. The contents of cellulose、hemicellulose and lgnin in the stalk of Triarrhena lutarioriparia L.Liu were increased, the change of sucrose content followed the order of decrease-increase-decrease along with growth stage. Compared with Diploid of Triarrhena lutarioriparia L.Liu, Autotetraploid of Triarrhena lutarioriparia L.Liu had the advantage of growth and physiological characteristics.2The effects of sucrose synthase activity and β-1,3-glucanase activity on the accumulation of cellulose were studied. Compared with Diploid of Triarrhena lutarioriparia L.Liu, sucrose synthsae activities and β-1,3-glucanase activities were higher in Autotetraploid of Triarrhena lutarioriparia L.Liu. During early growth stage, sucrose synthase activitiy and β-1,3-glucanase activity in the leaves rapidly increased, cellulose content in the stalk rapidly accumulated, sucrose content in the stalk rapidly decreased. Sucrose synthase activity and β-1,3-glucanase activity were significantly and positively correlated with cellulose content during the vegetative stage, repectively. It was suggested that sucrose synthase and β-1,3-glucanase activity had close correlation with cellulose accumulation and fiber quality in Triarrhena lutarioriparia L.Liu.3The effects of IAA, ABA content on the accumulation of cellulose were studied. During early growth stage, IAA content in leaves and the stalk increased, ABA content in leaves and the stalk decreased, cellulose content in the stalk rapidly accumulated, sucrose content in the stalk rapidly decreased. By correlation analysis, during early growth stage, IAA content were significantly and positively correlated with cellulose content, ABA content were significantly and negatively correlated with cellulose content. It was suggested that IAA,ABA content had close correlation with cellulose accumulation and fiber quality in Triarrhena lutarioriparia L.Liu.4A798bp of CesA gene sequence was cloned in Triarrhena lutarioriparia L.Liu, the sequences were bioinformatically analyzed. The results showed that the CesA gene sequences were conserved in the nucleotide and amino acid levels.
