Dynamics characteristics of macromolecules in the body fluids are an important basic theoretical research in biomedical applications. Basing on the application of the theory of Brownian motion as the background,, We make the theory research of silicon micro-spheres’ Brownian motion in the air and water in this papers, and analysis of micro-spheres’ Brownian motion characteristics in optical tweezers.First, comparison of the recent research results, we make an assay of velocity Boltzmann distribution and equipartition theorem of silicon micro-spheres; Second, we research on the mean square displacement and velocity autocorrelation function of single free particles in the air and water; We also research on the square root of the displacement as a function as the frequency of the potential well of particles in optical tweezers, and the mean square displacement and velocity autocorrelation function as a function as the time; Finally, we study the power spectrum of the silicon micro-spheres in optical potential wells. We analyse the factor of detector noise and other factors which effect on the results of measurement in the conditions of the constant laser power. We determine the state of motion to lay the foundation for extracting the Doppler motion information.
