6-amino penicillium alkanes acid (6-APA) is an important parent nucleus for the synthesis of beta-lactam antibiotics. The enzymatic methed for the producing6-APA has been becoming a research hot issue for its mild reaction conditions, less pollution and high conversion rate. In the enzymatic production of6-APA process, Penicillin G acylation enzyme is one of the important factor. Therefore, the research on immobilization of penicillin G acylase has a certain theoretical and practical significance.In this thesis, the main influencing factors of the immobilization process were studied, and the conclusion was obtained as followings:Firstly, the most important factor is the choice of the immobilization carrier. Through the comparative trial of three resin providers, the more suitable resin is the production from Xi’an Lanxiao new material Co. Ltd..Secondly, it is particularly important of external factors in the process of immobilization. We determined the enzyme ratio is the main factor through the orthogonal experiment analysis of the factors of the immobilization process. The influence sequence of various factors for Xi’an LanXiao resin is:enzyme ratio> temperature> pH> salt concentration.Thirdly, the optimum condition of the resin from Xi’an LanXiao new material Co. Ltd. was determined by orthogonal experiment, and the result was:immobilization temperature:26.5℃-27.5℃; pH:8; salt concentration:1.0mol/L; enzyme ratio:600U/g.
