Objective to establish the databases of mosquito rDNA-ITS and COI gene, so as to realize the molecular identification of mosquitoes, and establish the molecular basis of trace analysis based on COI gene.Method in2012, we collected the Chinese common mosquitoes (Culex pipiens pallens, Culex pipiens quinquefasciatus, Culex tritaeniorbynchus, Aedes albopictus, Anopheles sinensis, and Armigeres subbalbeatus) and identified them by the extraction of nucleic acid, gene amplification, sequencing and cloning plasmid vector, Multiple PCR, gel electrophoresis,sequence alignment and construction of phylogenetic tree.Result The full-length rDNA-ITS gene of Culex pipiens pallens, Culex quinquefasciatus, Culex tritaeniorbynchus, Aedes albopictus and Armigeres subalbatus can be cloned into plasmid vector, but the full-length rDNA-ITS of Anopheles sinensis can’t be amplified, only the rDNA-ITS2fragments were amplified. The fragment of Culex pipiens pallens and Culex quinquefasciatus is1273bp, the fragment of Aedes albopictus is1197bp,the fragment of Culex tritaeniorbynchus is995bp, the fragment of Armigeres subalbatus is1073bp.The different mosquitoes have the specific maps by agarose gel electrophoresis after multiplex PCR. We acquired9Culex pipiens group mtDNA-COI genes,which is in the regions of Lianyungang, Changshu, Zhangjiagang, Yancheng, Taizhou, Xuzhou, Nanjing, Nantong, Wuxi.Conclusion species identification by multiplex PCR on mosquito rDNA-ITS can realize those mosquitos; The COI gene of each regions in Jiangsu exist differences, We can get the souces infomation by the database.
