Flaxseed is one of the oil plants, it is emerging as one of the key sources ofphytoche-micals in the functional food. It is one of the important sources of α-linolenic acid,plant gum and lignans, and flaxseed is an essential source of high-quality protein, minerals,vitamins and soluble fiber. Many researchers of the world have paid much attention to SDGfor its good estrogenic activity. So it is necessary to study the extracting conditions for SDGproduction.In this paper, compared the content and yield of SDG were extracted by three types ofsolvent, and determined by HPLC: acetone is better than methanol and ethanol.The extractingcondition of acetone was further examined: It was that50%acetone extracted2times at50℃,2hours one time and the ratio of acetone to material was8:1(v/w).Used the fermentation by microbe was prepared for flaxseed, Flaxseed flour was addedwith2.5%of Asp.oryzae39and inoculated at room temperature about22-25℃for120hours.Then acetone solution was added into the fermented flaxseed flour to extract SDG. The yieldof crude extracts are21-22%and the yield of SDG is16-17mg·g-1, which is55%higher thanthat of no fermentation.The yield of SDG in defatted flax was20.92%, and the yield was6.6mg·g-1. The wholeextrcting water was mixed with3times volume of95%alcohol. And dried, the sedimentshad64%protein, the supernatant had SDG, and the yield was3.9mg·g-1.The use of ultrasonic extraction.The optimum time of extraction was2h. Compared withnon-ultrasonic extraction, the SDG extracts yield increased18%, and the SDG contentincreased10.5%. The yield of SDG was13.63mg.g-1, which increased30%thannon-ultrasonic treatment. Temperature has no significant effect on SDG extraction.
