Onconase, is a protein initially purified from extracts of Rana pipiens oocytes and early embryos based upon its anticancer activity both in vitro and in vivo. It is homologous to RNase A and is the smallest one in the superfamily.Onconase is the only enzyme of this class that reached clinical Ⅲ.Although ONC has effects with a series of tumor cells, it is still only used for the treatment of unresectable malignant mesothelioma. ONC is potent because of its low immunogenicity and high cytotoxicity.So our main purposes are using the genetic engineering methods to improve ONC to become even lower immunogenic and much more specific. We have already constructed three specific fusion proteins, of which only ONC-NSCLC shows us cytotoxicity as well as ONC.We also had constructed site-specific mutants. There are still many problems in the poccess of protein refolding, but we always try our best.
