Brachypodium distachyon is becoming a new model plant for genomics and biological of grass. The whole genome sequence of the diploid line Bd21has facilitated this process. But the transgenic research of Brachypodium distachyon is still weak. Agrobacterium-mediated transformation is the major method for the transformation of plants with its high transformation efficiency, exactlysegment transformationand low copy number of introduced genes. Here, we attend to establish and optimize the system of Agrobacterium-mediated transformation of Bd21. We found that MSB+MSB3medium is better to elevate transformation efficiency than1/2MS medium. And,one week of differentiation culture after screening can improve the efficiency of rooting from50%to90%. In this study, a complete system of Agrobacterium-mediated transformation of Bd21with higher efficiency was established, laying a foundation for genetic engineering. |
