Rotaviruses were classified into a genus of family Reoviridae, belonging to R otavirus. Rotavirus is a double-stranded RNA virus whose genome of11segments of double-stranded RNA composed of667-3302bp, which encoding six structural proteins (VP1-4, VP6, VP7), and5-6non-structural proteins (NSP1-5, part of the rotavirus NSP5gene encoding nonstructural proteins NSP6). According to antigen ic and gene sequence of VP6, rotavirus can be divided into seven groups (A-G) Group A, B, C rotavirus can both infect people and animals; While D-G group rotavirus only do animals. Group A rotavirus is one of the major cause of infants and young animals with severe acute gastroenteritis pathogenic.Rotavirus genome was consisted of0.6-3.5kb double-stranded RNA with varyi ng sizes. Different sizes revealed polyacrylamide gel electrophoresis migration rate, thus showing different bands. In early studies, according to the differences PAGE type analysis of rotavirus, which be divided into different group A rotavirus elec trophoretic type:long, short, ultra short and atypical rotavirus. Based on antigenic property of VP6, group A rotavirus was divided into different serotype subgroup s; Accordance with the prototype strain genomic hybridization results divided into genome hybridization type. In accordance with VP4and VP7antigenic serotypes group A rotavirus were divided into P and G serotypes going with a dual classif ication system, which15G and26P serotypes have been confirmed. With the dev elopment of molecular biology technology, a nucleotide-sequence-identity data of v p4and vp7were differentiated from P and G genotypes. Until by now, there are37genotypes and27G genotypes. NSP4gene identity analysis will be divided group A rotavirus into six different genotypes. Due to a variety of genetic reassor tment, rotavirus infection can occur between different strains simultaneously, the s ame P and G genotype strains and NSP4genotype may have a different genes. T herefore, Classification Working Group (RCWG) made whole-genome analysis clas sification system. The VP7-VP4-VP6-VP1-VP2-VP3-NSP1-NSP2-NSP3-NSP4-NSP5/6genes of rotavirus strains are described using the abbreviations Gx-P[x]-Ix-Rx-C x-Mx-Ax-Nx-Tx-Ex-Hx, respectively. So far at least27G (VP7),37P (VP4),16I (VP6).9R (VP1).9C (VP2).8M (VP3).16A (NSP1).9N (N SP2),12T (NSP3),14E (NSP4), andll H (NSP5/6) genotypes have been determined.Refer to the relevant literature and synthesis the primer, a rotavirus CC0812-1was isolated from an adult fece in Wuhan. RT-PCR primers were designed its entire11cloned gene segments, and complete sequencing. Multiple sequence align ments were aligned by using ClustalX2.1and DNAMAN, which compared CC0812-1with the other37P genotypes and27G genotypes reference strains of rota virus nucleotide and amino acid identity. Phylogenetic and molecular evolutionary analyses were constructed using the MEGA5.1software. It revealed that CC0812-1strain shared99.79%(99.61) nucleotide (amino acid) sequence identity of VP4gene with P[15] rotavirus strain LLR, which is first report detected P[15] ge netype GARV in human worldwide. Also CC0812-1strain shared99.15%(99.36) nucleotide (amino acid) sequence identity of VP7gene with G10rotavirus strain LLR, which comfirming CC0812-1and LLR are the same as a G10genetype RA RV.To confirm the relevance between the CC0812-1and LLR, we use RCWG r ecommended classification of whole genome CC0812-1are classified. Multiple seq uence alignments used ClustalX2.1and DNAMAN. Phylogenetic analyses were c onstructed using the MEGA5.1, phylogenetic trees were conducted by using the neighbor-joining method. Complete coding sequences of all segments for genetic a nalysis revealed a genomic constellation of this virus of G10-P[15]-I10-R2-C2-M2-A11-N2-T6-E2-H3. CC0812-1strain shared above99%homology identity with sh eep rotavirus Lamb-NT, Lamb-LLR, which proved that G10P[15] rotavirus strain CC0812-1is lapine rotavirus LLR vaccine-drived strain.
