The traditional paraffin section piece dyeing method has some disadvantages and limitations, such as the small production quantity, long production cycle, and high cost of time and energy, which is not suitable for mass production of section for teaching. The whole tissue H.E staining is a new method developed in recent years.In this study, we used self-made hematoxylin stain dye which contains15mg hematoxylin stain, 40mL distilled water, 600mg potassium alum, 5mg sodium periodate. The eosin stain contains250mg eosin R or B,50mL95% alcohol,5mL distilled water, a little acetic acid. Then by fixing, immersion, dip dye hematoxylin solution, staining, bluing, dehydration by gradient alcohol concentration, staining by eosin, alcoholic hydrocholric separations, dehydration by pure alcohol, transparent by xylene, embedding with paraffin and slicing. The sections obtained by this method are more uniform and colorful.Through the experiments, we found that the whole tissue slice staining is better than the traditional method in some aspects. It greatly simplified the production process, which can save time and energy for large producers. And it is appropriate for teaching and mass research sections of paraffin embedded tissue, and suitable for large batch parallel production. We carried out a deep discussion of the development evolution, adaptability in the production process, and advantages and disadvantages of two kinds of production methods.
