Radix pueraria was traditionally used both as medicine and food. It has good nutrition and be used as a functional food in our country. Radix pueraria whole powder is a kind of dehydrated product isolated from Radix pueraria. The powder is nutritional and instant which contain all of the material except peel. The coefficient of utilization to radix pueraria can as high as80%to90%. Radix pueraria is rich in fibers, and cello-oligosaccharide make up from the fibers by hydrolyze. The cello-oligosaccharide is a kind of nature, safety and novel functional oligosaccharide, which can not be used both by people and animal when they are digesting but which can help the Bacillus bifidus to get increase. This powder is a new kind of production and the research on the preparation of the powder has high economic signification and academic value. The task of this paper was to study the preparation of radix pueraria whole powder containing cello-oligosaccharide on the basis of previous researches.1. Some properties of polyphenol oxidase (PPO) and different effects of chemical inhibitors on PPO were studied in this experiment. The results showed that the optimum pH value of PPO was4.4, and the optimum temperature of PPO was55℃. PPO was deactivated under the heating treatment of90~100℃for1~3minutes, compound reagent made of1.5%sodium chloride,0.75%citric acid and0.3%calcium chloride had a strong inhibition on PPO activity of radix pueraria. During the procession, the compound reagent was used to keep the original color of radix pueraria products.2. After the treatment of super micro-milling, both the basic composition and functional composition of radix pueraria whole powder varying with particle size were studied. The results demonstrated that the particle size was considerably reduced after the treatment of super micro-milling, the content of starch, fat, protein, reducing sugar, and ash were not changed, but the total dietary fiber (TDF) content reduced, the soluble dietary fiber (SDF) content increased, the insoluble dietary fiber (IDF) redused. The dissolution rate of flavonoid increased obviously.3. The cello-oligosaccharides were prepared by enzymatic method, and the composition and content of the sample through high performance anion-exchange chromatography were analyzed. The result showed that β-glucanase was better than cellulase to produce cello-oligosaccharides that the main hydrolytic components were glucose, cellobiose and cellotriose. Optimization of hydrolysis conditions was investigated by orthogonal test, and effects of pH, amount of β-glucanase, reaction temperature and reaction time were all tested. The optimal preparation catalyzed by β-glucanase was appeared at the follow conditions: pH at5.0, β-glucanase at3U/g, reaction temperature at50℃and reaction time at2h, respectively.4. The results from the dryness experiments showed that the dispersity and colour of the product were not good by use of extrusion dryness, and the retention rate of flavonoid was low. The lyophilization was better than extrusion dryness to produce Radix pueraria whole powder.
