This thesis deals with the study of fluorescence spectra of1-Naphthalene boronic acidand the investigation of the influences of experimental conditions on fluorescence spectra.And some researches are conducted on the reaction between1-Naphthalene boronic acid andsome monosaccharides under different experimental conditions. Further, a high performanceliquid chromatographic method by using FL detector is proposed for the determination ofD-Glucose, D-Fructose, Arabitol and Sorbitol respectively.This thesis falls into three parts:Part1. In this part fluorescence spectra of1-Naphthalene boronic acid are studied. Andan inspection has been made on the influence of ionic strength, acidity, solvent, surfactanteffects on spectral properties and an investigation has also been conducted on the linearrelationship between fluorescence intensity and concentration. The fluorescence quantumyields of1-Naphthalene boronic acid are finally measured to be0.16.Part2. A research is undertaken in terms of the fluorescence spectra changes of1-Naphthalene boronic acid reacting with D-Fructose, D-.Glucose, D-Galactose, D-Mannoseand Sorbitol in different experimental conditions. A finding has turned out that under thealkaline condition(pH=11), the fluorescence intensity of1-Naphthalene boronic acid isstrengthened after adding monosaccharides, the order of which goes this way: Sorbitol>D-Fructose> D-Galactose> D-Mannose> D-.Glucose. Ultimately the reason leading to theboost of the fluorescence intensity in different degrees is studied.Part3. This part is centered on the establishment of method dealing with a fluorescencedetector1-naphthalene boric acid by high performance liquid chromatography (HPLC-FL)with post-column derivatization and determination method of D-glucose, D-fructose, Arabitoland sorbitol in Arabia. The chromatographic conditions and derivatization conditions are thusoptimized, the results of which demonstrate that four kinds of sugar can be separated underthese conditions, good linearity, recovery rate of drink and honey samples are between88%-96%and78%-103%, the relative standard deviation (RSD) is less than3%(n=6). Themethod proves to be sensitive, accurate, and suitable for the determination of the content ofmonosaccharides.
