In order to realize HPLC analysis of complex samples, a novel, simple and miniature two dimensional carbon fiber sample pretreatment column has been developed and it was applied in pre-separation of different polar components in the complex plant extract. In which, the first and second dimensional pre-columns were simply made by carbon fibers and activated carbon fibers, respectively. The complex chemical mixtures were roughly separated to non-polar and polar components using first dimensional pre-column, and then the low polar and high polar chemicals were separated with second dimensional pre-column. The three fractions were directly provided to HPLC analysis without any other sample pretreatment processes after concentration. The reliability and sensitivity of the method was evaluated by the analysis of a mixture of13standard compounds applying UV detection. Under the optimized conditions, recoveries of target compounds ranged from75.4%to93.9%, relative standard deviation (RSD%) ranged below than14.6%, correlation coefficients were above0.99, limits of quantitation for these targets ranged from0.10ng to11.0ng. Only0.2g of sample and less than1mL solvent were used in the whole sample pretreatment processes. The technique was greatly reduced the amount of organic solvent and sample, and labor to compare with traditional sample pretreatment. In addition, comparison of directly HPLC-UV analysis of plant extract, it obtains more components and high resolution. Finally, the method was applied to determine of real Chinese medicine samples. The results show that this method is simple, rapid, economic, easy to operate, high throughput, make plant sample total analysis come true and it has widely application prospect in total analysis of complex samples.
