Drug sustained-release drug delivery systems reduce the frequency ofadministration, to maintain the plasma concentration, improve drug efficacy, reduce sideeffects of drugs, more and more researchers began this system as a new drug researchdirection. Silk fibroin as a natural protein, has also been found that can be sustained andcontrolled release medication polymer materials. Curcuma oil is a kind of anti-virusanti-tumor traditional Chinese medicine, are likely to play a greater role in the increasingincidence of neoplastic diseases.In this paper, a sustained-release system for the design premise cleverly combinessilk fibroin Curcuma oil use, the use of self-assembly properties of the silk fibroinprepared sustained release capabilities the silk fibroin embedding Curcuma oil releasemicrospheres.First established a method for the determination of the purpose of drug Curcuma oil;determination of different concentrations of solvent, select the hydrochloric acid solutioncontaining1.2%Tween-80in simulated gastric environment, containing1.2%Tween-80in PBS simulated intestinal environment. And then through the the salt dissolved dialysisprepared the key carrier material salt-soluble silk fibroin, after select salt-soluble silkfibroin to prepare fibroin embedded Curcuma oil release microspheres. Select theuniform design and orthogonal design were investigated stirring speed, emulsiondenaturation time, the amount of emulsifier, silk fibroin concentration, the amount ofanhydrous ethanol and other conditions, was successfully prepared uniform particle size,low pungent odor average drug20.51%, average encapsulation efficiency was96.07%fibroin embedded Curcuma oil release microspheres by infrared spectroscopy, laserparticle size analyzer, scanning electron microscopy to determine the successfulpreparation of the microspheres. Release microspheres security test, hemolysis results4hdid not happen hemolysis, might have developed into injection; the microspheres LD50=1015.860mg/kg, higher microspheres security. Finally, by the MTT method microspherestoxicity to mouse fibroblast L-929cells, to inhibit tumor cell growth in vitro activity ofthe microspheres by Hela tumor cell lines was evaluated, the results show blank fibroinmicrospheres cells growth promoting effect of sustained-release microspheres lesstoxicity to normal cells, significantly inhibit the growth of tumor cells, with asustained-release effect. The results showed that the prepared pharmaceutical properties of silk fibroinembedded Curcuma oil release microspheres can be developed as injections and oralsustained-release formulations, such a preparation process can be used for similar drugswith the nature of the Curcuma oil, provides for new formulations research directions.
