Purification And Biological Activity Evaluation Of Flavonoids From The Chloroform Raffinate Phase Of Taxus Mairei Leaves

Recently Taxus were mainly studied on Taxol,10-DAB and other taxanes, rather than nontaxane compounds such as flavonoids. Suspending the material liquor by chloroform was an important procedure to get out taxanes from Taxus extracts. The raffinate phase was treated as barren liquor, which made the material utilization ratio reduced. As to reduce the manufacturing cost and protect biological resources of Taxus, the technology of flavonoids isolation and purification from the chloroform raffinate phase of Taxus was optimized.With the recovery ratio of flavonoids as the indicator, the technology of flavonoids isolation and purification from the chloroform raffinate phase of Taxus was examined. Refer to the single factor experiments, orthogonal design experiment was performed with4significant factors (Sample concentration, sample flow rate, eluent concentration, and eluent flow rate) considered. After preparative isolation with AB-8macroporous resin and polyamide resin in series, the content of the flavonoids in the final products was increased to53.23%, and the total recovery ratio was58.74%.In addition, the biological activities of the flavonoids in the final products of the chloroform raffinate. phase of Taxus were determined. The antioxidant activity experiments indicated that the purified products performed elimination effects on the DPPH free radical. The antimicrobial activity experiments indicated that the purified products performed significant inhibitory effect on4kinds of test bacteria and broad spectrum inhibitory effect on4kinds of test fungus.
