The aims of this study were to screen high-yield cellulase strains and to improve theproduct enzyme conditions. Based on seven trichoderma viride strains, seed medium andfermentation medium processing, the enzyme activity of the C1, CX, CBand degradationrate of cellulose and lignin were assayed to screen good strains with higher enzymeactivity and degradation rates; By antagonism experiment of Trichoderma viride andwhite-rotfungi, two bacteria should be mixed fermentation for the step two todegradation of straw; Trichoderma viride and white-rot fungus mixed fermentationconditions was optimized by the proportion of straw and bran, the water content inmedium, total inoculum of mixed bacterias, inoculation ratio of mixed bacterias,respectively; Adopted two levels in vitro digestion test, the in vitro digestibility of thefermented straw was evaluated and the results were as follows:(1) CMC enzyme and filter paper enzyme activity from Trichoderma viride L4C andBS were the highest when straw as the main carbon source, and the activity of them was2438.4U/mL,2438.4U/mL,715.4U/mL and658.8U/mL, respectively. When wheatused as the main carbon source, CMC enzyme and filter paper enzyme activity fromtrichoderma viride L4C and BS became the highest, and the activity of which was2854.0U/mL,3237.7U/mL880.5U/mL and865.7U/mL, respectively.(2) The test to use mixed fermentation of Trichoderma viride and white-rot fungishowed that the fermentation time was six days, and the best combination was O6-H10,namely white-rot fungi inoculated for6days, then inoculated trichoderma viride for10days.(3) The optimization condition of Trichoderma viride L4C, BS and white-rot fungion straw medium were as follows: the proportion of straw and gluten for7:3,6:4, thewater content in medium for55%,45%, the total inoculum of mixed bacterias for15%,25%,the inoculation ratio of mixed bacterias all for3:2. Undering the bestfermentation conditions, the hemicellulose, cellulose, lignin degradation rate of mushroom and Trichoderma viride L4C were36.4%,41.6%and48.1%,respectively.namely that were1.5times,1.22times and2.5times, respectively. The degradation rateof bacteria fermentation; mushroom and Trichoderma viride BS were31.4%,31.5%and52.9%, respectively, namely that are1.3times,1.05times and2.75times, respectively.The optimization condition of Trichoderma viride BS, L4C and white-rot fungi on wheatmedium were as follows: the proportion of wheat and gluten was7:3, and the watercontent in medium was55%, the total inoculum of mixed bacterias was15%,and theinoculation ratio of mixed bacterias was3:2.(4) In vitro two levels digestion test showed that dry matter digestibility of mixedbacterias fermentation was reach to40%~60%, that was1.79~2.82times as high ascontrol.
