Environmental-responsive polymer materials can adapt to surrounding environments,regulate transport of ions and molecules, change wettability and adhesion of differentspecies on external stimuli, or convert chemical and biochemical signals into optical,electrical, thermal and mechanical signals, and vice versa. These materials are playing anincreasingly important part in a diverse range of applications, such as drug delivery,diagnostics, tissue engineering and ‘smart’ optical systems, as well as biosensors,microelectromechanical systems, coatings and textiles. Here, a series of ‘smart’ nanogelsbased on poly(ethylene glycol) methyl ether methacrylate (OEGMA) were constructed,then the characteristics, the drug loading and relaese behaviors of drug-loaded nanogelswere studied. At last, the inhibition effection on human liver cancer cells, Bel7402, wasstudied.P(OEGMA-ss-MEO2MA) shorted for POM P(OEGMA-ss-MEO2MA-ss-Am)shorted for POMA and P(OEGMA-ss-MEO2MA-ss-MSD shorted for POMS aresynthesized through precipitation polymerization, these three kinds of nanogels are allredox-sensitive. Then doxorubicin (DOX) was loaded into nanogels with three methods:physical adsorption, high-speed shear and hydrophobic force driven by TEA. Themaximum drug-loading efficiencies determined by centrifugal ultrafiltration was98.9%,and the highest drug-loading level was17.8wt%. The drug release behaviors andinhibition effection on tumor cells were performed as follow. The practicability ofapplying OEGMA based muti-sensitive nanogels on cancer targeted drug administrationwas investigated preliminarilyIn brief, OEGMA based nanogels have high biocompatibility, biodegradability anddrug loading efficiency. POMS nanogel has high drug loading level, and it’s volume phasetransition behaviors can change with different enviroment. So POMS nanogel is expectedto be an effective anti-cancer drugs carrier.
