Alkylated chitosan which had good film-forming ability was the product of chitosan modification.The alkalization system with microwave heating was used to prepare the alkylated chitosan by modification. Then, the ethyl chitosan, butyl chitosan, lauryl chitosan, cetyl chitosan were prepared. The structure of products were determined by FT-IR and X-ray. The antibacterial properties of alkylated chitosan were investigated. It was known that only the butyl chitosan has inhibition on Gram-positive Salmonella-Staphylococcus aureus. So butyl chitosan as raw materials, the film-forming process of butyl chitosan and its application in drug delivery were examined in detail.The forming process of the butyl chitosan film was selected, the production process of excellent butyl chitosan film were as follows:3%butyl chitosan solution was prepared by butyl chitosan dissolved in4%acetic acid solution,which forming film original solution, then it was transferred to the glass petri dish and cast into membranes. After it was dried at40℃, the dry film was soaked in a6%sodium hydroxide solution for3h, then it was washed clean to the neutral. The film of butyl chitosan was obtained. The film obtained in this condition with the breaking strength was53.5Mpa, the elongation at break was13.7%, the average swelling rate of the film was53.34%.The drug was wrapped with membrane by the double-membrane sandwich method. VC as a model drug, the effect of drug delivery was investigated at pH of3.5,3.6,3.7,3.8. At a pH of3.5. VC drugs wrapped in the membrane would be gradually slow-released with the degradation of butyl chitosan film. The releasing of the drug was controlled by changing the sustained-release environment. At pH3.5,the drug release rate achieved80%after6h.At pH3.6, the drug release rate reached80%after8h.These provided a basis to design the corresponding drug release film depending on the different drug properties and action site in the future.
