This research used peanut shell as raw material, mainly studied the process for isolating xylan from peanut shell with alkali and ultrasonic wave. A Response Surface Methodology (RSM) was used for optimization of isolation of xylan. Then the xylanase hydrolysis of xylan for xylo-oligosaccharide is optimized using RSMThe response surface methodology (RSM) was employed to optimize the four most significant factors:sodium hydroxide dose, liquid-solid ratio, reaction time and reaction temperature in xylan isolation with alkali. The model predicted a maximum xylan extraction ratio of38.2871%under the optimal conditions of sodium hydroxide (12.47%), liquid-solid ratio (1:14.93), reaction time (2.07h) and reaction temperature (69.82℃). Finally,the optimal technological conditions were correctted to the odium hydroxide (12.5%), liquid-solid ratio (1:15), reaction time (2h) and reaction temperature (70℃).The final result is38.08%The response surface methodology (RSM) was employed to optimize the four most significant factors:sodium hydroxide dose, ultrasonic power, reaction time and reaction temperature in xylan isolation with ultrasonic wave. The model predicted a maximum xylan extraction ratio of45.9931%under the optimal conditions of sodium hydroxide (8.49%), ultrasonic power (137.23W), reaction time (79.92min) and reaction temperature (61.48℃). Finally,the optimal technological conditions were correctted to the odium hydroxide (8.5%), ultrasonic power (137W), reaction time (80min) and reaction temperature (62℃).The final result is46.21%Xylanase added amount, hydrolysis temperature, substrate concentration and hydrolysis time is the four major factors of the xylan hydrolysis reactions.The optimal xylan hydrolysis conditions are xylanase added amount (1000IU/ml), hydrolysis temperature (55℃), Substrate concentration (6%) and hydrolysis time (5h).The average degree of polymerization of xylo-oligosaccharide is3.1and the yield of xylo-oligosaccharide is30.01%Because of the xyol-oligosaccharides solution Browning after xylanase hydrolysis,so the modified activated carbon was used to decolorating xylanase hydrolysis solution in order to obtion high purity xyol-oligosaccharides. Activated carbon added amount, reaction temperature, pH and reaction time is the four major factors of the activated carbon decolorizing reactions.The optimal activated carbon decolorating reaction conditions are activated carbon amount (12%), reaction temperature (90℃), pH (3.0) and reaction time ( 80min).The Decoloring rate is44.3%and the loss of xylo-oligosaccharide solution is20%.
