Studies On Evaluation Techniques Of Chloramphenicol ELISA Test Kit And Melamine Colloidal Gold Test Strip

As banned additives, chloramphenicol and melamine were important content of the food safety inspection, and the immunological methods has been widely used because it had the advantages of high-throughput, low prices, convenient operation, the national monitoring program of veterinary drug residues has already regarded ELISA as a reference method. But the kits in market were uneven in quality, and the users, producers, regulators were eager to have standardized and scientific evaluation techniques to distinguish good and bad.We combined the foreign evaluation techniques and the evaluation requirements of domestic ministry of agriculture, we formed the evaluation system whose patameters were ensitivity, specificity, accuracy, reproducibility, the detection limit, stability and inter-assay coefficients of variation and established the evaluation methods of the ELISA test kits and colloidal gold test strips. In addition, we chose several chloramphenicol ELISA test kits and colloidal gold test strips which market sales volume were large as objects of study.The test results show that the corresponding detection limits of kit A which for chicken muscle and shrimp flesh were0.077,0.128ng/ml, and the corresponding detection limits of kit B which for chicken muscle, shrimp flesh, Chicken liver, milk, honey and royal jelly were0.0212,0.0232,0.0179,0.00817,0.0213,0.0255ng/ml ng/ml, so they conformed the limits which referred to the" monitoring program of veterinary drug residues in animal and animal products of2012". The corresponding detection limits of kit D which for raw milk and powdered milk were200ng/mL,2μg/g, so they met the limitation requirement in the relevant national standard.This paper provided evalution method for the related kits, and it could help users to select products, and help producers to make quality control scheme, and provided the basis of law enforcement for regulators.
