The use of preconcentration steps based on phase separation bythe technique shows high efficacy for quantitative extraction,operative simplicity and safety. Other methodologies have beendeveloped, such as membrane extraction and solid-phaseextraction (SPE), solid-phase microextraction (SPME) etc. Cloudpoint extraction as a new extraction technology has been usedsuccessfully for the preconcentration of species of widely differingcharacter and nature due to no exposure to more dangerous andtoxic organic solvents when it is compared to other traditionalextraction procedures, such as metal ions, proteins and otherbiomaterials, or organic compounds of strongly differing polarity. InCPE, separation of two phases, the surfactant-rich phase with theisolated analytes and the aqueous supernatant phase with thesurfactant close to critical micelle concentration, is causedmainly by the influence of temperature. The compounds separationtechnology based on magnetic materials has received considerableattention in recent years. It provides great potential for applicationsin cell separation, drug delivery and enzyme immobilization. In this paper, a new combined methodology of CPE and fluorimetricdetermination of drug has been developed. The content of thispaper are as follows:1. The Principle cloud point extraction and magnetic separationtechniques was introduced. Detailed study of the research progresswhich is about cloud point extraction and magnetic extractioncombined with variouty of detection equipment inorganic and organiccompounds.2. A cloud-point extraction process combining HPLC using mixedmicelle of the anionic surfactant sodium dodecyl sulfate (SDS) andthe nonionic surfactant polyoxyethylene(7.5)nonylphenylether(PONPE7.5) to extract three fluoroquinolone antimicrobial agentsciprofloxacin, norfloxacin and lomefloxacin from aqueous mediawas investigated.At the optimum conditions, the rectilinearcalibration graphs were obtained in the concentration rangeenrofloxacin1-1500ng mL-1, enoxacin0.5-1500ng mL-1, Pefloxacin0.5~1500ng mL-1, of the limits of detection was0.2ng mL1.Inpractice, the method is simple, fast, green.3. A novel two-step extraction technique combining cloud pointextraction (CPE) with dispersive micro-solid phase extraction ispresented in this work. The method involves initial extraction of thetarget analytes about Rh6G by CPE in the micelles of anionicsurfactant surfactant (SDBS), then highly hydrophobic diatomitesupport magnetic nanoparticles (MNPs) are used to retrieve themicellar phase. Rich phase is determined by fluorescencespectrophotometry During the experiment to optimize the extractionof various parameters. At the optimum conditions,The average recoveries of tap water and lake water samples was97.6%~100.8%,the detection limit was0.006ng mL-1,4. It established a two-step method that is the use of diatomitesupport magnetic materials combined with the condensed phasecloud point extraction for Chinese medicine about three isoquinoline.Then rich phase is determined with high performance liquidchromatography combining UV detector. At the optimum conditions,the rectilinear calibration graphs were obtained in the concentrationrange0.1~1000ng mL1of three isoquinoline with a detection limitwas0.03ng mL1, respectively. The proposed method has theadvantage of a high sensitivity, environment-friendly and reproducibleIt is applied for the determination of the drug in urine and serumsamples in the pharmacokinetics.
