Recently, huge quantity of hypersaline organic wastewater discharged every year in China, which was great harmful to the ecological environment and became the big problem in environmental engineering field. Moderate halophiles possessing strong metabolism ability widely existed in the habitats with extended range of salinity, which had great importance to the biotreatment of hypersaline effluents. In this study, hypersaline phenol wastewater was degraded in the Sequencing Batch Reactor by the moderately halophilic bacterium which can survive in a large rang of salinity after domestication. After59d of acclimation, the concentration of NaCl could be120g/L in the SBR1, and included two dominant genera: Halomonas and Pseudomonas. The SBR have different performance on the degradation of phenol in different concentration of NaCl; the phenol concentration could be1600mg/L after82days of acclimation at the existing of50g/L NaCl; the phenol concentration could be1200mg/L after84d of acclimation at the existing of80g/l NaCl; The dominant genus changed from Brachybacterium and Halomonas to Pseudomonas and Alcaligenes after the domestication.PCR detection of key genotypes suggested the positive of the existing of phenol hydroxylase(Lph), catechol1,2-dioxygenase (C12O) and the ortho-pathway or catechol2,3-dioxygenase (C23O) both in the original sludge and the ultimate sludge. RT-PCR was employed to indicate that the amount of total bacterial changed from1.00×1011copies/g-sludge to7.22×105copies/g-sludge while the amount of Lph changed from2.30×108copies/g-sludge to7.78×103copies/g-sludge when the NaCl concentration increased from30g/L to120g/L; the amount of total bacterial changed from9.50×108copies/g-sludge to1.94×107copies/g-sludge while the amount of Lph changed from1.20×107copies/g-sludge to3.49×106copies/g-sludge when the phenol concentration increased from400mg/L to1200mg/L.
