Hydroxyanthraquinone derivatives, which is an antimicrobial, antiphlogistic andanticancer thing, is the mainly effective component of rheum officinale. Phyllosticta isan endophytic mycelium of Rheum officinale. The phyllosticta of rheum officinalehave the ability to produce hydroxyanthraquinone derivatives. Hydroxyanthraquinonederivatives have some biological activities, such as antibacterial and anti tumor. Itdivides into two kinds of existing form in plants, which is free type and glycosidetype.This article studies on extraction method of anthraquinones from fermentationliquor of phyllosticta. Using the orthogonal experimental design method to find outthe best technological conditions as follows: fermentation days for20d, organicextraction solvent for chloroform and liquid ratio for50:1. Under such experimentalconditions, the average yield of anthraquinones was23.96mg/g. Usingspectrophotometer to detect, establish a quantitative measurement method ofanthraquinones from fermentation liquor of phyllosticta. The standard curve wasy=23.00+0.015x, R2=0.997. The content of rhein shows a good linearity relationshipbetween0.001mg/mL and0.025mg/mL. Verification experiments have beendesigned and made, which shows good precision, reproducibility, stability and highsample recovery rate. Using IR spectrum and NMR methods to identify the structureof the purification, determined that the compound was rhein (1,8-dyhydroxy-3-carboxylic acid anthraquinone). Establish a high performance liquidchromatography (HPLC) method for quantitative analysis of anthraquinones fromfermentation liquor of phyllosticta. Study on the bioactivities of the extraction fromfermentation liquor of phyllosticta by using filter and MTT methods to textantibacterial and anti-inflammatory activities. The antibacterial assay shows thatchloroform extracts can obviously restrain the four indicator bacteria, especially forthe salmonella paratyphi. The anti-inflammatory assay shows that LPS can induceRAW264.7cells release NO, and rhein have an obvious inhibitory effect on such cells when the safe dose of rhein is15mg/L. The antioxidant activity of rhein is examinedusing various antioxidant assays, such as reducing power, DPPH assay and lipidperoxidation. The antioxidant activity assay shows the rhein is isolated as the one ofmajor flavonoids from fermentation liquor of phyllosticta with antioxidation.
