The purpose of this study was to increase the production of adenosine deaminase and the density of E.coli with the improvement of the nutrition、the temperature and the process control.The comparison of different temperatures shown that:37℃was better than30℃for the production of ADA, while the complex medium was better than synthetic medium. But at37℃,in the complex medium the bacteria was easy to be prematuredecay, while at30℃or in the synthetic medium the growth of the E.coli was better. So we tried to reduce the growth rate of the bacteria at the early stage in the complex medium at37℃. Under this control,the density of E.coli and the production of the adenosine deaminase were all increased. This was very important for the large scale production.Through the study of the water content and the RNA content in the bacteria,we found that the different water contents reflected different growth conditions, the fast decrease of the water content may be the result of the prematuredecay. RNA content and the changes of it were different under different nutrition conditions. The maximum of the RNA content was higher in the rich medium of complex medium than the synthetic medium, so the RNA content can be an index of the nutrition. The change tendency of the RNA content had connection with growth rate. The reduction of RNA content after induction may have connection with the growth state of the cells. So through the detection of the RNA content, we could see the growth condition of the bacteria.
