To further clarify the anti-tumor effect of active proteins from Syngnathus, the extraction protocol of anti-tumor active protein from Syngnathus was optimized and established using active effect-oriented method.Anti-tumor active proteins from Trachyrhamphus serratus, Syngnathus acus Linnaeus, Solenognathus hardwickii, Syngnathoides biaculeatus were also obtained with the extraction protocol, and proteins from this four officinal Syngnathus had no significant difference on the anti-tumor effect in vitro and had the same molecular weight.The results showed that the anti-tumor active protein from Syngnathus could induce apoptosis in cancer cells, which proved by flow cytometry, and morphologic assessment. And it could enhanced the inhibition effect of MTX strongly. The toxic and animal experiments revealed that the anti-tumor active protein could enhance the immune system and inhibit the growth of tumor in S180 tumor-bearing mice.These results would provide a merit for further development and reasonably utilization of Syngnathus and halobios resources.
