Chinese mitten crab (Eriocheir sinensis) which is delicious and nutritious is one of the important economic crabs in China. With the increases of market demand, the farming scale of E. sinensis expands unceasingly. However, the phenomenon of sex precocity which usually occurred in cultural process brings great losses, because the precocious crab is easy to die. The normal developing and maturing of male crab in the reproduction process of E.sinensis is one of the important influence factors. Studying its reproductive physiological and biochemical characteristics such as spermatophores, sperms, mortality, acrosomal reaction rate as well as sperm protein is of important theoretical significance and practical significance for analysing sperm quality and germplasm characteristics as well as realizing the reproductive index of precocial crab.The experimental materials were normal and precocious male E. sinensis. Firstly, using paraffin section technology we contrasted the tissue differences of testis between normal precocious male crab; by counting the spermatophore and sperms and measuring diameter of sperm, we got the relevant statistic data of sperm; we got the qualitative data by detecting the mortality of sperms and the rate of sperm acrosome reaction to analysis the difference between normal precocious crab. Then we analysised the zymogram and enzyme activity of LDH between normal precocious crab by the way of native polyacrylamide gel electrophoresis stained with LDH activity dyeing liquid. At last, using two-dimensional electrophoresis technology, we contrasted the proteome between normal precocious crab to find out the different protein.The result showed that the testis tissue in precocious crab had a higher maturity than that in normal crab. The number of spermatophores and sperms in normal crab were respectively1.35times and1.32times as that in precocious crab. The mortality of sperms in normal and precocious crab were respectively1.9%and3.42%, and the rate of sperm acrosome reaction were respectively93%and87%. The enzyme activity of LDH and LDH-X in normal crab sperm were higher than that in precocious crab.281and296protein spots were acquired respectively from2-DE of sperm protein in normal and precocious crab. The results showed that the precocious crab was short of normal crab on reproductive capacity and the quality of offspring gametes, some differences in sperm proteome existed in this two species.
