Objective: To developed, optimized and compared the colloidal goldimmunochromatography assays (GICA) and fluorescence quenching ICA fordetecting furaltadone metabolites3-amino-5-morpholinomethyl-2-oxazolidinone(AMOZ).Method: The monoclonal antibodies against CPAMOZ labeled by nanoparticle wereused as probes spraying on binding pads. CPAMOZ-BSA and goat anti-mouse IgGdispensed as the test line and control line on nitrocellulose membrane, respectively.The binding pad and nitrocellulose membrane was assembled gather with sample padand absorption pad to form GICA strip. The sensitive, specificity, stability of GICAwas tested, after optimized reacting conditions. Fluorescent particles were mixed intotest line and control line was changed into fluorescent particles only to developfluorescence quenching ICA. The sensitivity, detecting range and cross-reaction wereverified after optimized conditions, the above dates of two kinds of ICA werecompared.Result: The CPAMOZ-BSA was synthesized successfully. The GICA andfluorescence quenching ICA were developed. The sensitivities of them were10ppband0.625ppb respectively. The detecting range of fluorescence quenching ICA was0.3-10ppb. Both of two ICA had low cross reaction with AHD and SEM and SEMand their derivative of4-CBA.Conclusion: In this study, two methods of ICA for detecting AMOZ were establishedsuccessfully. Compared with GICA, fluorescence quenching ICA improved thesensitivity, broadened detecting range, and realized quantitative determination.Meanwhile, the fluorescence quenching ICA retained the advantages of GICA-fast,high specificity, low cost-which only required simple equipments. This method issuitable for large high-throughput screening at the grass-roots level.
