In order to understand the infection and strains of Echinococcus granulosus and determine its genotype of Echinococcus granulosus in Inner Mongolia.This research was conducted the epidemiological investigation of Echinococcus granulosus which included2,938sheep and113cows fed in different area of Inner Mongolia (Bameng, Ximeng, Wumeng, Erdos),meanwhile identified cysts which removed from six patients through liver surgery. At the same time, we collected226canines fecal from different areas in Inner Mongolia(Bameng, Ximeng, Wumeng),and canine echinococcus antigen was detected by ELISA.The cysts which obtain through the epidemiological investigation were cloned and sequenced partial sequence of the CO1gene and ND1gene by PCR amplification,and the the sequence of measured CO1gene was compared with the sequence of CO1gene of Xinjiang strain of Echinococcus granulosus which loged in the GenBank,it may understand the variation of Echinococcus granulosus.Simultaneously, the genotypes of Echinococcus granulosus were determined through the sequence of measured ND1gene. The results were showed as following:1. There were only one adult sheep that infected the disease of Echinococcus granulosus in the2938sheep;113cows were not infected the disease of Echinococcus granulosus;the cysts that removed from six patients through liver surgery were Echinococcus granulosus.2.The detected results of226canine echinococcus antigen were shown that the simples positive rate of Wulatezhongqi was20%, the simples positive rate of Siziwangqi was3.8%, the simples positive rate of Dongwuqi was13.8%, the simples positive rate of Xilinhot was8%, and the simples positive rate of Xiwuqi was3.3%.3.The sequence length of CO1gene which was sheep strain from Xiwuqi and people strains from Xilinhot of Echinococcus granulosus were936bp.Compared with the sequence of CO1gene of Xinjiang strain of Echinococcus granulosus which loged in the GenBank, its corresponding region exists6variable sites in the sheep strain of Xiwuqi and Xinjiang strain, the homology was99.3%; its corresponding region exists3variable sites in the people strains of Xilinhot and Xinjiang strain, the homology was98.6%.4.The sheep strain from Xiwuqi and the people strains from Xilinhot of Echinococc-us granulosus were G1genotypes by DNAStar5.0software.The research results showed that the infection rate of Echinococcus granulosus was very low. The sequence of CO1gene which was sheep strain from Xiwuqi and people strains from Xilinhot of Echinococcus granulosus and the sequence of CO1gene of Xinjiang strain of Echinococcus granulosus which loged in the GenBank had a high homology.The sheep strain and people strains of Echinococcus granulosus were G1genotypes through epidemiological investigation from Ximeng areas in Inner Mongolia.This study results provided theoretical basis for developing generic diagnostic kit of Echinococcus granulosus disease.
