| Infectious Bovine Rhinotracheitis Virus can cause bovine multiple tissues infection especially respiratory, in recently year,this disease is prevail and cause enormous economic losses. This study isolate and culture bovine mononuclear macrophages in vitro and detect phagocytic function, NO secretion of bovine mononuclear macrophages after infected with IBRV at6,12,24,48h,respectively and analyse the effect of IBRV infection on immunology function of bovine mononuclear macrophages at cellular or molecular level.The result of Phagocytosis of chicken red blood cell test shows that the Phagocytic function of mononuclear macrophages decrease after infected with IBRV (P<0.05). Apoptosis of mononuclear macrophages after infected with IBRV was not founded in agarose gel electrophoresis and fluorescence microscope detection;the effect of IBRV on NO secretion of bovine mononuclear macrophages was detected by Griess method and the results shows that the NO secretion higher than control group after infected with IBRV(P<0.05),and reach to the highest level at24h after poison;The dynamic changes of expression of Surface molecules MHCIã€MHCIIã€CD14ã€CDlla of mononuclear macrophages were detected by flow cytometry, the results shows that the relationship between IBRV infection and the expression of CD14and CDlla which associated with anti-infection immunity is Positive regulation but Negative regulation with MHCI and MHCII expression which related with antigen presentation.That is to say, bovine mononuclear macrophages infected by IBRV can influence the expression level of cell surface molecule.The result proved that, in short time,the Phagocytosis function and antigen presentation function were decrease, Immune regulation function was affected of bovine mononuclear macrophages after infecting with IBRV and provided theory evidence for further study the Pathogenic mechanism of IBRV. |
