The Effects Of Puerarin To Growth Hormone And Prolactin Levels And Receptor Expressions In Mice

To illustrate the effects of Puerarin towards growth hormone (GH) and prolactin’s (PRL) levels of mice serum and on growth hormone receptors (GHR) and prolactin receptor’s (PRLR) gene expression’s dynamic changes in breast tissue, this test divides240mice of21days old into four groups, as the control group, low dose of Puerarin, middle dose of Puerarin and high dose of Puerarin, each group has60mice, administered once every other day. The mice of control group is given0.2ml of distilled water, and the low dose group given0.2ml of0.5mg/ml Puerarin solution, the dose group given0.2ml of1.0mg/ml Puerarin solution, and high dose group is given0.2ml of1.5mg/ml Puerarin solution. And respectively in the40,46,52,58,64,70day, the mice of each group are killed by cervical and then collect the blood and take the breast, with the method of radioimmunoassay and real-time quantitative RT-PCR to detect the growth hormone, prolactin’s levels in serum and growth hormone receptors, prolactin receptor’s expression in breast tissue.The results showed:1. Puerarin tends to improve growth hormone (GH) level of mice serum, low, medium and high dose of puerarin group can all enhance the growth hormone (GH) level. But in70days the high dose of puerarin is to reduce the growth hormone’s concentration.2. To the different days group of mice, low, medium and high dose of puerarin group all are to show less effect toward prolactin level of mice serum.3. Compared with the control group, the low, medium and high dose of puerarin group benefit the expression of the growth hormone receptors in40,46,64,70days while restrain the expression of growth hormone receptors in52,58days. For the different days mice, the low dose of doesn’t have notable effect to the expression of the growth hormone receptors, the medium dose can improve the expression of growth hormone receptors with growth of the age and the high dose have the remarkable the improve.4. Compared with the control group, the low, medium and high dose of puerarin group benefit the expression of the prolactin receptors in40,46,64,70days while restrain the expression of prolactin receptors in52,58days. In term of the different days of mice, the low, medium, and high dose of puerarin enhance the expression of the prolactin receptors in58days.
