Ghrelin is a new endogenous brain-gut peptide consist of28kinds of amino aci,which was found by Japanese scientist Kojima in1999,it is a natural ligand of growth hormone secretagogue receptor.Its variety functions depend on its wide distribution in the body. At present many people are concerned of the hot point.Corpus luteum was transformed from theca lutein cells and granulosa lutein cells after ovulation. There were a lot of changes on biology and molecular biology in the formation process of corpus luteum, and the mRNA expression of P450scc and3β-HSD, which were required for progesterone synthesis, and they were reduced after the LH surge before ovulation, but the mRNA expression of those above and the enzyme activity increased in the stage of after ovulation and luteinization, and progesterone production increases, meanwhile, there have conformance in the synthesis of StAR and P4, therefor, P4, P450scc,3β-HSD, and StAR was the sign on the formation of the corpus luteum.For further confirm the effect of Ghrelin in Mongolian sheep luteinizing granulosa cells, this study investigate secretion of P4after adding different concentrations hormone to the cultured sheep granulosa cells in virto. In order to make the granulosa cells finish the differentiation to lutein cells in virto.The results showed that the group with2.5IU/ml LH-. FSH and E2(lug/mL) for24h had an increased progesterone secretion than control(P<0.05). Then real-time PCR was used to detect Ihe levels of mRNA expression of the equivalent luteinizing genes. Compared with the control group, mRNA expression of the equivalent luteinizing genes was markedly up-regulated.Then adding Ghrelin into luteinizing granulosa cells,100ng/ml Ghrelin markedly inhibited gene expression of3β-HSD、 CYP11、StAR.20ng/ml Ghrelin promoted the expression of gene than100ng/ml Ghrelin,but inhibited the expression of gene with blank control. The results contribute to further research on the effect of formation and degeneration of the corpus luteum.
