The Chinese yam(Dioscorea opposita Thunb.) tuber was studied as the material after harvest and then stored at constant temperature of4℃. First, the best extraction conditions of tuber polysaccharides and mucilage polysaccharide was investigated respectively by the orthogonal test method. Second, yam tuber polysaccharide and mucilage polysaccharide was extracted respectively by the best extraction condition on different varieties, storage period and parts. Third, the best index of different yam variety, storage period and parts in polysaccharides physiological function was investigated respectively by the orthogonal test design and then the correlation was analysed between each index and these two knids of polysaccharides content respectively. Fourth, Chinese yam tuber polysaccharide and mucilage polysaccharide physiological function was investigated and compared under different concentrations of tuber polysaccharides and mucilage polysaccharide. The experimental results were as follows:1. The best yam tuber polysaccharide extraction condition was studied by the single factor test and four factors three levels of orthogonal test. Extraction temperature was90℃, pH value was7.5, feed liquid ratio was1:5, extracting time was3h. The best yam mucilage polysaccharide extraction condition was as follows:ethanol final concentration was80%, centrifugal time was10min, beating time was90s, centrifugal speed was3500r/min.2. Chinese yam tuber polysaccharides and mucilage polysaccharide physiological function was determinated in different varieties, storage period and part respectively by orthogonal test. The results were all the HNTGJ29varieties, tuber central, the first storage period of yam with the different impact order of each index.3. Both of Chinese yam tuber polysaccharides and mucilage polysaccharide content showed significant positive correlation with its reduction force absorbance value, O2.-free radicals to the clearance rate,·OH free radicals to the clearance rate, DPPH·free radicals to the clearance rate, NO2-·free radicals to the clearance rate, linoleic acid suppression force, the alpha amylase inhibition rate respectively.4. Mucilage polysaccharide solution of reducing power stronger than tuber polysaccharide solution; mucilage polysaccharide solution to O2-·free radicals removal was stronger than tuber polysaccharide solution; tuber polysaccharide solution to·OH radical removal was stronger than mucilage polysaccharide; tuber DPPH·free radicals polysaccharide solution to remove was stronger than mucilage polysaccharide solution; nucilage polysaccharide solution to NO2-·free radicals removal was stronger than tuber polysaccharide solution; mucilage polysaccharide solution of linoleic acid oxidation nhibition was stronger than tuber polysaccharide solution; mucilage polysaccharide solution with alpha amylase inhibitory rate was higher than tuber polysaccharide solution nhibition rate.
