Swine Fever Vaccine,Live(Spleen and lymph tissue origin)is short for Classical Swine FeverVaccine,Live(rabbit origin), the characteristic of Classical Swine Fever vaccine are strong immunogenicityand good immune effect, but the production process of tissue which caused larger production cost is rathercumbersome, low work efficiency, large labor intensity. Without affecting the quality of products under thepremise, comparison test was done between the traditional production method and improved productionmethod by reducing time of measuring temperature and increasing the number of harvest tissue, Study ontissue preservation method,and the best protective agent and temperature were obtained by preservationmethod.It takes21times to measure temperature during the whole production process. Through comparing theresult of statistic,it was showed that the results of measuring temperature at48h and72h was consistentwith regulations, the vaccine that produced by two different method showed that the potency test result wasno difference.the improved method can greatly reduce the labor intensity,meanwhile, the measures will helpdecrease the cost of production and productivity has increased as a result.The CSFV virus of Spleen and lymph tissue origin was derived from the tissue of rabbit which has feverreactivity, if other tissue which contained CSFV could be found in rabbit, it would increase the production.The tissue of Liver、Spleen、mesenteric lymph nodes、submandibular lymph nodes were collected and thevirus content was detected, the results indicated that the virus content in liver was low and submandibularlymph nodes was hard to harvest and easy to be Contaminated,both of these could not meet productionrequirements, Spleen and mesenteric lymph nodes are the best tissue for production.In the test normal saline、Hank’s、milk protective agent and heat resistance protective agent of classicalswine fever were used for protecting the virus and stored at different temperature, retention period test weredone by detected the temperature of rabbit,the result showed that milk protective agent retention periodlasted longer at-15℃and was the most economic and efficient.The results of research can increase productivity and to achieve the aim of reducing the productioncost.
