| In order to research the effects of substitution and changes of HMW-GS to eachsubunit relative content and quality of wheat in the same genetic background.In thisstudy, obtained by continuous selective backcross six generation (BC6) respectively.three near-isogenic lines (NILs) of Longfu3(subunits5+10were located at Glu-D1locus while subunits2*,1and N were located at Glu-A1locus) and four near-isogeniclines (NILs) of Longmai19(Glu-D1locus were5+10or2+12subunits, Glu-A3/Gli-A1locus were Glu-A3c/Gli-A1m or Glu-A3e/Gli-A1? subunits)as experimental material,through reversed-phase high-performance liquid chromatography (RP-HPLC) anddifferent quality detection method to analysis the effect of HMW-GS substitution andthe fluctuation of quality indicators of wheat and each subunit expression changes,discusses the relationship between the subunits quality and quantity.Conclusions are asfollows:The genetic background of Glu-D1loci5+10subunit, the two years qualityanalysis results of the NILs Longfu3which contains HMW-GS2*,1and N coded atGlu-A1loci showed that the dry gluten content, wet gluten content of NILs Longfu3had no significant difference statistically, the protein content, gluten index and Zelenysedimentation value had large significant difference. Farinograph parameters ofdevelopment time,stability time and breakdown time of NILs had highly significant; Inthe seven main parameters of the mixograph, in addition to the peak bandwidth and8min bandwidth no differences, the other parameters differences significant;From theAlveograph parameters we could find that P value,L value and W value of NILs werevery significant; Extensogram parameters showed that the differences of the maximumresistance and areas. When2*and1subunit substitution N subunit,settling time, breaktime, W values, the maximum resistance and tensile area have the greatest impact,2*subunit substitution were increased by71.7%,70.7%,39.8%,50.8%and50.3%onthese indicators.overall contribution of Glu-A1subunit on the quality effects size were2*>1>N. While the relative content of HMW-GS each subunit was determined byRP-HPLC, Longfu3wheat varieties NILs Glu-A1locus the size of relative content were1>2*>N,but1subunit relative content only large1%than2*subunit, the contentis substantially the same,and the relative contents in the other locus had largedifference.Quality analysis of Longmai19wheat varieties of4kinds (2+12,Glu-A3c/Gli-A1?,2+12,Glu-A3e/Gli-A1m,5+10,Glu-A3c/Gli-A1?,5+10,Glu-A3e/Gli-A1m)NILs showed that,4kinds of NILs of flour protein content had no significantdifference, there were significant differences in the wet gluten content, dry glutencontent, gluten index, Zeleny sedimentation value and Zeleny sedimentation value/drygluten. Farinograph parameters of stability time and breakdown time of4kinds ofNILs had highly significant; The seven main parameters of mixograph in the left peakbandwidth, peak bandwidth without difference, other parameters were significantlydifferent; Longmai19wheat variety4kinds of NILs of Alveograph parameters showsthat significantly differences in P value and L value, reached extremely significantlevel in W value;4NILs extensograph parameters in the extensibility of no obviousdifference, significant difference in the maximum tensile resistance and tensile area.From look on the whole, the4kinds of NILs on the quality of the contributions ofD613-5>D626-5>D613-2>D626-2, analysis shows that the effect of5+10and Glu-A3csubunit of gluten index, Zeleny sedimentation value, stability time, breakdown time, Wvalue, the maximum tensile resistance and tensile area of these quality indexes havethe largest contribution, D613lines and D626lines in5+10than2+12subunits in themaximum tensile resistance are increased by100.6%and94.2%, in the tensile areawere increased by79.2%and95%, the quality of wheat contribution of size5+10>2+12, Glu-A3c>Glu-A3e. By RP-HPLC measures HMW-GS and LMW-GSexpression of Longmai19wheat varieties4near-isogenic lines and calculate therelative content, relative content of5+10subunit and2+12subunit is basically thesame, containing the Glu-A3c subunit as the genetic background of the lines of relativecontent of the LMW-GS is greater than Glu-A3e subunit as the genetic background ofthe lines.These results indicate that2*subunit and1subunitã€5+10and2+12subunits isbasically the same in quantity by RP-HPLC, the quality indicators show that thecontribution of2*subunit is better than that of the quality of1subunit,5+10subunit isbetter than2+12subunit,5+10contribution to the quality is double the2*subunit contribution to the quality. Introduce quality subunit, when the same or similar subunitexpression, should select large quality contribution subunit,5+10quality subunitshould be widely introduced strong gluten wheat genetics and breeding, while in thechoice of high-quality subunits should give full consideration to the changes in therelationship between quality and quantity.RP-HPLC can accurately detect the quantityof different subunits and calculate the relative contents, NILs can exclude theinterference of the genetic background, so research the relationship of the expressionyield of different subunits of wheat and quality use of RP-HPLC with NILs as materialmore accurately. |
