| Rab proteins are members of the largest subfamily in Ras superfamily that playimportant role during the process of vesicle transport as molecular switches. Rab5islocated in coated vesicles and early endosomes and responsible for regulatingendocytic internalization and early endosome fusion. Rab7is distributed in the lateendosome, regulating the early endosome to late endosome transfer process.There are11putative members of Rab family in Magnaporthe grisea genomeDatabase. Sequence analysis showed MoYpt51(MGG06241), MoYpt52(MGG01185) and MoYpt7(MGG08144) and their homologous proteins includedthe conserved G-domain and highly variable N-terminal and C-terminal. G domainstructure has five highly conserved amino acid sequences RabF1-F5and fourcharacteristic sequences RabSF1-4in Rab protein family. Phylogenetic analysisshowed that they are homologous to N.crassa and S.cerevisia, so choose S.cerevisia asa model fungi in Interolog approach.Through many large protein interaction databases searching and literaturesupport, we received hundreds of interacting proteins and interactors. By informatictechnology and high-performance graphics platform, the protein interaction networksare constructed. The networks were separated into several biological modules. Thefuntions of them contained involving in protein degradation of the ubiquitin pathway,relating to vesicle transport, and playing an important role in the assembly and modifyprocess of proteins and nucleic acids. The topology structure analysis found theprotein interaction networks compared with random network had obvious scale freenetwork and small world properties. These networks had certain module clusteringcharacteristics. Subcellular location showed that most interacting proteins located inthe cytoplasm and the plasma membrane, that are the same as target proteins.According to domain analysis results, we selected MGG07137andMGG04143for yeast two hybrid validation. Predicting protein MGG07137andactive MoYpt7have a certain degree of interaction. In consideration of a largenumber of literature, it may be related to the inactive MoYpt7protein; MGG04143did not interact with MoYpt5and MoYpt7, so this protein may not be their directinteractions. From its domain interacting with Ras family speculated that it mightregulate other Rab family proteins rather than directly effect on the MoYpt5andMoYpt7, or needed some modification to interact target proteins and this can be achieved by other experiments such as co-immuno precipitation and GST pull-downto confirm. |
