With the material of resistant ‘6-12-4’ and susceptible ‘6-12-2’ grape plants, theexpression of the candidate resistance genes were studied by qRT-PCR and the whole genometranscription were studied by Digital Gene Expression based on RNA sequencing afterpowdery mildew inoculation. The primary results were shown as follow:1. The results of qRT-PCR indicated that the expression of12candidate resistance geneswhich were obtained from the cDNA library were all up-regulated compared to the controlafter induced by powdery mildew. The expression variation ranged from0.32folds (36h) to11.93folds (24h) in resistant grape ‘6-12-4’ while from0.31folds (12h) to3.79folds (6h)in susceptible grape ‘6-12-2’. The gene expression variation according to the hours postinoculation can be classified into single peak, double peak and linary type.2. Compared to susceptible grape, the relative expression of4candidate genes of theresistance grape which showed significant up-regulated expression, and they were0054(24h,9.50folds),0132(12h,2.47folds),1007(24h,4.14folds) and1045(6h,3.98flods).3. According to the largest relative expression in the resistance and susceptible grape,combined with the bioinformation, the candidate resistance gene0054,1007and1044weresignificantly induced and supposed to be resistance gene. All of them were localized in thecaryon, cytoplasm and cytomembrane, and showed no specific distribution in cells.4. By DGE analysis, the remarkablely differential expressed genes at6,24,48hour postinoculation were acquired.At6hour post inoculation, there were72differentially expressed genes in resistantstrain compared to non-inoculation.41of them were up-regulated, and the expression variedfrom17.41to2.22folds of control. Among them12genes were grouped into molecularfunction and15in cellular component and15genes about biological process. Besides, thefunctions of24genes were unknown. Meanwhile,31genes were down-regulated, and theexpression varied from2.11to15.69folds of non-inoculation. Among them8genes wererelated to molecular function and11genes about cellular component and12genes aboutbiological process. In addition, there were16genes whose functions kept unknown. At24hour post inoculation, there were8differentially expressed genes and all of them wereup-regulated. The expression varied from2.28to14.55folds. Among them2genes were related to molecular function and2genes to cellular component and3genes about biologicalprocess. Besides, the functions of5genes were unknown. At48hour post inoculation, therewere7differentially expressed genes.6of them displayed up-regulated, and expressionvaried from2.53to14.36folds. Among them2genes were involved molecular function and3genes related to cellular component and3genes about biological process. Beyond that, thefunctions of4genes were unknown. Meanwhile one gene whose function was unknowndisplayed down-regulated and its expression varied range was3.64folds.
