Stripe rust, caused by the fungus Puccinia striiformis f. sp. Tritici(PST), is the majordisease of wheat worldwide. This disease can spread widely by air and cause serious damageto wheat production. Planting resistant cultivars is the most economical, effective, andenvironment-friendly way to control the disease.But, for the reason of kinds of races of PSTand variations, a new variation may overcome the resistance of wheat cultivars. Theinheritance background of the resistant wheat cultivars growing in China is narrow and thiscan accelerate the pathogen directional selection, the resistant wheat cultivars may becomesusceptible quickly. So, it is significant to explore the existing resistance resources, discoverthe new resources and increase the diversity of wheat resistance resources for sustainablecontrol of wheat stripe rust.In this research, the source of the resistant cultivar is abundant. The field resistance atadult stage of110resistant cultivars to stripe rust was tested in artificial nursery at Yanglingand in natural nursery at Tianshui for several successive years, the results show that they arehighly resistant to stripe rust at adult stage. During2010-2012, for confirming their adultresistance, we continue to test the resistance at Yangling and Tianshui. To know more aboutthe genotype and genetic characteristics of the resistant cultivars, Using12prevalent isolatesof PST to investigate its resistance spectrum at seedling stage, and the resistant resources wasdetected with the known DNA markers linked to rust resistance genes Yr5, Yr9, Yr10, Yr15,Yr17, Yr18and Yr26.In this study, Qinnong142has good disease resistance and ideal agronomic traits. Forbetter studying and using the resistance sources in wheat breeding program, it is important tofind out the characterization and inheritance of resistance to stripe rust. The resistanceinheritance of Qinlong142was investigated with F1plants, F2plants, F2:3lines.Achievedresults are as follows:1. The phenotype of resistant resources to stripe rust: At adult stage, the wheat cultivar03155-2-12and3323-5-2-3H105display highly resistance to stripe rust during2010-2011.But, in2012,they are susceptible to stripe rust. The other108wheat cultivars display highlyresistance to stripe rust during2010-2012. At seedling stage,19wheat cultivars are highly ormoderate resistant to all PST isolates. They have non-race special resistance gene, the adult resistance of them can’t be confirmed;88wheat cultivars are resistant to a part of the PSTisolates, but susceptible to the rest of the PST isolates. They have race special resistance gene.3wheat cultivars(FHB143USA, H84CIMMYT132and Shan757) are susceptible to allisolates, they have adult resistance gene.2. The results of molecular marker detection and gene postulation: Combined with theresistance spectrum and results of molecular marker detection of wheat monogenic lines andresistant resources, analyzing the resistance gene. The results shows that:9wheat cultivarshave Yr5, which account for8.2%;27wheat cultivars have Yr9, which account for24.5%;3wheat cultivars have Yr10, which account for2.7%;3wheat cultivars have Yr15, whichaccount for2.7%;5wheat cultivars have Yr17, which account for4.5%; no wheat cultivarhave Yr18;5wheat cultivars have Yr26, which account for4.5%;65wheat cultivars haveunknown resistance gene which need more research to confirm them.3. Inheritance at adult plant in a wheat cultivar Qinnong142: At adult stage, After yearsof research, confirmed that Qinnong142is highly resistant to stripe rust; At seedling stage,Qinnong142is highly or moderate resistant to all PST Isolates except for isolate CH42. Theresult shows that Qinnong142is typical adult resistance resource.
